Possibly yes. Possibly no. If this is not a standard tooltip (in other
words, if the last tooltip hotkey doesn't read the information you're
expecting), then it's possible a script could be written to watch for
those windows, and when discovered, speak their information. Of course,
that's all theory until I can get a demo or something in front of me to try.
HotSpot is more about creating mouse macro commands: move the mouse
here, do some action or actions. With the upcoming HotSpot, you won't
even need to have the mouse in the right place; you'll be able to have
the HotSpot search for a string (even with specific attributes) and move
the mouse to it automatically, kind of like the current Mouse find
comment (only automatic), and then perform any number of actions. Plus,
with the ability to chain HotSpots together, your potential will be
unlimited.

The Audible sample button is a great use for the hot spot script. I have
it defined so when I press enter on a book so I can read the publisher's
summary, I can press alt-shift-s and hear the sample. the hot key routes
the mouse to the sample button and left clicks. Then my sample is played.
Another use for the hot spot script is in Quicken 2009 where I can press
a hot key and reconcile, update, or do various other tasks which
normally require me to move the mouse pointer to the specific part of
the screen I want.
