Dr. Charles Scouten addressed this issue regarding changes of brain due to different perfusion fixation techniques. He has an image showing the difference between a fresh brain, one pressure perfused with sucrose followed by 4% paraformaldehyde/glutaraldehyde, and the last one with gravity flow perfusion using the same materials. My thought is that if you see shrinkage of the material depending on perfusion fixation methods, you will likely see it with immersion fixation.
