Hi,
     Soak the coverslip off and let it sit in xylene long enough to make
absolutely sure all the mounting media is dissolved. Then starting from
100% ETOH rehydrate to water then buffer. And start the IHC as usual.
     There is no need to destain. The ethanol will remove the eosin and you
are likely to counterstain with hematoxylin anyway. Make sure you do repeat
the counterstain as well since the hematoxylin will probably bleach out if
not entirely, certainly some.
