1) Do my users have to enable it on their clients before I enable it on the server (currently clients only connect to my network for sending mail) or does this have to be set simultaneously at the client and at the server?
You have to coordinate it, because requiring SMTP AUTH slams the door on their sending mail if they haven't held the door open with SMTP AUTH in their clients.

Set an hour and date when you start requiring SMTP AUTH and have them get with the program by then.

2) If I can set SMTP Authentication on the server first, will those clients that connect directly to my network require setting this option prior to being able to send mail?
no, they get through with relay for addresses, but I recommend that try to get everybody to use relay for addresses, so you have a trace of every sender.
