Yes.

SMTPD32  receives  incoming  messages  over the SMTP protocol, whether
destined  for  local or remote domains. Messages submitted using other
protocols,  such  as  those  from  Web  Messaging,  do  not go through
SMTPD32.

In 7.1x and lower versions, yes. In 8.x, SMTP32 isn't even used at all
unless  there's  a  message filter such as Declude installed, and even
then  it's  just  a  tiny shim to get stuff to Queue Mgr, which is the
replacement delivery app.
