We have installed Diskeeper on our Email Server, and schedule a defrag
during non-peak times (from 10pm to 4am). The defrag runs every 2 hours
during this time.

After 3 months I ran a scan on that server and the file and disk
defragmentation is still always kept below 1%. That's pretty impressive in
my opinion.

and to keep defragging to reasonable times, the spool should on a dedicated 
partition, and the mailbox storage on its own partition.
