Yes, up to three instances of the "nameserver" keyword can be included in
the file.

Of course, this will only be of use to applications running on the router.

Applications running on hosts behind the router will refer to their own
resolv.conf (or equivalent), and if those are configured to refer to
dnscache running on the router, then the dnscache configuration will
determine the upstream hosts.  This comes preconfigured with the root
servers, which are about as reliable as it gets. (dnscache ignores
resolv.conf.)
