Once your message is written, shift tab to the signature field which is to the right of the from field, so you shift tab once and then make sure your signature is selected as you need to put a name to call your signature when you are in mail preferences. I hope you are going in to mail preferences with command comma and writing your signature that way rather than system preferences.
