Hi,

For testing, what is wrong, could You provide sample tile rendered with
png and png256 format (same region, only different format).
You could create ticket:
http://trac.mapnik.org/report
and add attachment.

Meantime, You can try using "png256:m=h" instead of "png256", see if it
helps.
