How can I specify multiple instances from a single config file? Is it unnecessary to do that?

I don't think you can - but you could make an amended copy of /etc/ init.d/memcached-init and point it a different copy of /usr/share/ memcached/scripts/start-memcached, which you can point at a different config file, then just start it as a separate service.
