When setiing up fill the upper buffer chamber first with around 200ml buffer and check for leaks before filling the lower chamber (600ml). Replace the XCell Surelock gaskets if they are leaking , Invitrogen code: EI9021.
The recommended voltage for NUPAGE Bis-Tris gels is 200V or 150-175V for 9 and 17 well gels.
