There are two methods to calculate absorption coefficients at 280 nm if 
the amino acid composition of the protein is known:
S. J. Perkins: Eur. J. Biochem. 157 (1986) 169-180
S. C. Gill & P. H. von Hippel: Anal. Biochem. 182 (1989) 319-326
Once you have epsilon, you only need to apply Lambert-Beer's law.
