Hi I wanted the check the concentration of purified recombinant protein at 280 nm which is at native form. To measure the concentration of it, do i need to unfold the protein or native form would be fine., which one will give best results. If any of the aromatic acids are not exposed to the solvent, do they participate in absorbance?
