I think a bigger buffer, better write speed, and at least a 2.5" LCD are givens - that's just what's off-the-shelf for technology today. ISO 50 (and, presumably, a higher megapixel count) depend on what's available in sensor technology. I expect more megapixels because that's where I see the market pressure - I'd guess 10MP by that time. Beyond that, I don't know what to expect. Features I don't expect to see are the aperture simulator lever, or a so-called full frame sensor. I doubt if we'll even see a lower crop factor such as 1.3; I believe Pentax are sticking with a 1.5 crop factor for the moment. Much as I'd like to believe that the next DSLR will be a camera of the quality of an LX, I don't think that will happen - anything much more than an MZ-S (or even the *ist-D itself) just isn't probable. I don't think Pentax can sell a $2500 K-mount body. More to the point, I don't think Pentax believe that, either; I'd be extremely surprised to see a recommended release price even as high as $1500.
