I don't think you can do this with Win32::Service but you can with Win32::Daemon by Dave Roth. The modules "ConfigureService" routine allows you to pass in a hash reference of configuration settings of which one hash key can be "start_type" and can have a value of either SERVICE_AUTO_START, SERVICE_DEMAND_START or SERVICE_DISABLED.
