Yes, rewrite postfix to use a DB as a backend rather than a file system. This will be a major redesign and not just a plugin. No one else is working on such a project, so you're on your own.
If you require your mail queue to be stored in MySql, you'll need to find a different MTA because postfix just can't do that.
