No, iTunes doesn't read the metadata either. iTunes does not support
metadata in wave files at all. But if you would loose your iTunes
database for some reason, then it is possible to recover it by using my
application (if your wave files are tagged of course). It has a feature
called initialize iTunes database. This feature reads the metadata from
the wave files and sets the iTunes information about that file
accordingly. 
There is a readme file available at the provided download link above
that explains the various features in more detail.
