Hello,
Does anyone has a list of category can share that is used for IHC CRO
comparison? I want to make a table to do comparison after talking to
multiple CROs and I don't want to miss something.  Just wondering if anyone
did this before and can share your table or list?

[Histonet] NIS Elements

We used to use Image-Pro Premier to capture and analyze images. Does anyone
use NIS Elements with Nikon to do image capture and analyze? How do you
feel about it? If you used both of them, could you let me know what you

[Histonet] rodent heart tissue harvesting

Hello histonet,

My coworker wants me to ask experts here the best way to use Potassium
Chloride during rodent heart harvesting. Could any one share your
procedure? We searched around on internet but not much help.

If there's any way other than Potassium Chloride to relax heart muscle,
please advice too.

Thanks to all who responded to my question regarding PAS staining!
We got much better staining after following you guys suggestion:
a. we purchased reagent from American Mastertech
b. we used 1% periodic acid
c. we rinse slides under warm water after Schiff's step.

[Histonet] PAS staining

Hello,

We tried to do PAS staining on kidney. I attached my picture here. I am
looking for the staining results that has more reddish as showing in the
second picture. So I can do quantification better. We purchased  Schiff's
reagent from Sigma freshly. Could anybody teach us  how can I can the
staining like that?

[Histonet] How to fix frozen section?

Hello,
 
I will receive some frozen tissue sections that are not fixed. I rarely do IHC 
on frozen sections. I searched some articles about fixation. Some use 50/50 of 
acetone/alcohol and some use only alcohol. Could you please recommend a 
fixation method for me?

[Histonet] neutrophil antibody on rat FFP section

I was asked to perform neutrophil IHC on rat FFP sections. I searched around 
and found most are for mouse tissue. There is one from Serotec mouse anti rat 
but it does not work on paraffin section. 
 
Could anybody recommend a good antibody?

[Histonet] IHC blocking for liver tissue

Hello,
 
I have a question that what is best way to block endougenous biotin and HRP in 
liver or kidney tissue. The method I am using has to use avidin-biotin and HRP 
reactions. This is fine on heart tissue.  I used biotin blocking kit from DAKO 
or Vector. Also I used 1% H2O2. But liver tissue still show background stain.
 
Could you teach me a better way to block or is there other reason cause 
background?

[Histonet] To detect injected mouse antibody inside mouse tissue

Hello,
 
I have a question regarding detecting injected mouse antibody in mouse tissue 
by IHC.
Now I have mouse liver paraffin section. This mouse was injected mouse 
antibody. I was asked to locate this antibody. This is a mouse chimeric 
antibody. Variable regions from human antibody was combined with mouse IgG2a 
heavy chain and kappa light chain constant regions. I am thinking using rabbit 
anti-human IgG, F(ab) fragment specific. Do you think it's doable or you have 
any good suggestion? When this mouse antibody bind to antigen inside liver 
through this human variable region, my F(ab) fragment specific antibody can 
still bind to it?
