[ccp4bb] off-topic: protein losing FAD during purification

Dear All (those dealing with wetlab stuff..),

While purifying a FAD containing protein we lose part of the FAD (on the gel 
filtration we clearly see two bands corresponding to holoprotein and free FAD).

We obtain crystals but diffracting to only about 4 A despite their beautiful 
look. Our hypothesis is that the crystals contain a population of molecules 
with and without FAD (?).

The questions are:

1) how to keep FAD bound to the protein during purification and crystallization?

2) how to completely remove FAD from the protein?

A 3 year PhD fellowship is available to a suitable candidate for one of the 
following projects involving molecular biology and structural biology 
techniques:

Molecular characterization of the bio synthetic pathway of amylovoran, a 
pathogenicity factor in Erwinia amylovora

Biomolecular study of iron uptake by desferrioxamine in the plant pathogen 
Erwinia amylovora

the work will involve cloning of genes, protein expression, purification and 
crystallization and structure determination by X-ray crystallography. Proteins 
will be characterized using in solution techniques including NMR available 
through collaboration with other laboratories.

[ccp4bb] Enterokinase cleavage of His-tagged proteins

I decided to use a LIC vector with enterokinase cleavage site for His tag 
removal. Of course I did not check the length of the linker (about 45 aa) nor 
if the enterokinase cleavage site would be the best choice

 

Do you know where I could purchase a decent quality with reasonable price 
enterokinase to cleave mgs of his-tagged proteins for crystallization?

I need to buy a Linux workstation to run crystallographic software and graphics 
like ccp4, mosflm, coot., etc.,

Could you please suggest me a good combination of hardware and  which linux 
operating system to install (ubuntu?)? I can spend about 1500€

Technology evolves so fast that I really want to be up to date not to be 
already late!
