[Histonet] Can I use no water benzoyl peroxide from PolySciences in my MMA mixture?

I try to avoid CaCl drying step in making MMA infiltration and embedding 
solutions. Please help me MMA experts. Many lab use benzoyl peroxide which has 
30% water in it. And go through CaCl afterwards. 
Another question, why some lab use Butanol and toluene before coverslip? Why 
use Cellosolve to do deplastify? I think xylene and ethanol are cheaper and 
less toxic, is it true?

Thank you to everyone who sent a template.  This is very helpful.  I do
need clarfication on the 10 cases that are required for validation.  Do you
include the 10 cases on this same form, or do you have something separate
for that?  These look ideal for optimization.  From what I have read
online, 10 cases need to be run after optimization has been completed.
When validating these 10 cases, accuracy, sensitivity, specificity and
reproducibility need to be monitored.  Are all of you using separate form
to capture this data and approval from the Pathologists?  Maybe I am going
overboard, but I want to make sure that I have everything covered.  We are
bringing a new platform into our lab in 2 weeks, and I want to have
everything ready to go.

[Histonet] doing histology in an academic research setting

Hi everyone,


I have been doing histology professionally for the past 5 years in an
academic research lab setting (and an additional 5 years for my own grad
studies).  I take pride in the samples I have processed, cut, stained, and
I love being able to troubleshoot any challenges or questions that the lab
encounters. While I have this feeling for the actual work churned out, I
can’t help but feel left out in lab meetings when actual data are presented
and discussed. It makes me feel I don’t do enough. I guess what I’ve
struggled with is that I do want to be a part of doing data procurement and
analysis, but with the amount of work routine histology requires there’s
not enough time in the day to also do microscope work and count for data.
For those who are in a similar setting, do you share the same feeling?

My apologies in advance if this is a silly thing to share/ask, but it has
been bugging me for some time now.

[Histonet] Antibodies to fungus

I think I asked this question a few years ago, but I'm going to ask again 


DAKO used to sell monoclonal antibodies to Aspergillus (clone Mab-WF-AF-1) and 
Rhizomucor (clone Mab-WSSA-RA-1).  Unfortunately, they discontinued these 
reagents several years ago.  Does anyone know if these antibodies are still 
available through another vendor?  Thank you.

two if not three different validation means (PCR, micro, serology, etc.)
Anyone out there actually doing this?
Just so you know, I've spoken with a few suppliers on control slides and they 
do not do this type of validation on their control material (surprise, 
surprise!)

Denise,
When selecting infectious agent positive controls, I also select cases
which have been shown to be positive by other testing means (EM,
Bacteriology, Virology, PCR).

In addition, I run a cross-reactivity validation with my antibody on other
infectious agent positive control slides to make sure my antibody is
specific for the infectious agent and doesn't cross-react with other
viruses/bacteria, etc.  Results go into a table that is stored along with
the initial antibody validation/optimization logsheet(s).

When validating tissue antigens/tumor markers, I use the obvious normal
anatomic cell/tissue control for that antigen.  My anatomic pathologist
advisor signs off on the final optimization for all antibodies before the
IHC test can be put into public use.
