We use whatever is the narrowest range for the reagents stored in there. [Histonet] Prostate Needle Bx
How do others process prostate needle bx's - what kind of cassettes do you use? Do you use sponges? Do you wrap the tissue in lens paper? Do you process them on a biopsy (short) run? Our bx's always seem to be less than optimal, but we do not have problems with other small bx's. I have discovered a wonderful cassette from Cancer Diagnostics, it is called the Autocassette and has an oval concave cavity with no corners for small pieces of tissue to get caught in and the solutions pass through it very nicely. I use it for small research samples, like shrimp gills similar in size to the prostate biopsies, and I lose nothing. I use the yellow ones, VB1005 but they have other colors and also some with smaller grid. Highly recommend them. [Histonet] MMA Mounting Issue with Haupts Adhesive
Hi all, I had a question about how to properly mount MMA sectioned slides using Haupt's (Dorn and Hart Microedge). My protocol is as follows: 1. Stick Glass Slide into 50mL Falcon Tube with Haupts solution (1:1 50% EtOH:50% Haupts). 2. Rotate the tube to ensure entire coating of glass slide. 3. Let glass slide dry in fume hood for about 10minutes. 4. Remove cut microtome sections (range: 50-5microns). 5. Place cut sections in 70%EtOH to soften the plastic and unroll. (sections are coming out rolled up like scrolls) 6. Remove a section from the 70%EtOH using forceps and try unrolling it with a needle on a glass slide. 7. Once the section(s) is stretched on the coated glass slide, cover with saran wrap. 8. Cover the wrapped slide with coarse filter paper and use a small seam roller to roll the sections on the glass slide as flat as possible. This will also help remove excess 70%EtOH. 9. Remove the filter paper and stack glass slides in a slide press or similar device. Place tongue depressors or small pieces of wood equivalent to the slide length that are about 1cm thick at the end of the glass slide stack. 10. Let slides dry over night at room temperature in press. 11. Remove the slides from the press and unwrap the saran wrap to expose your adhered sample. Questions/Comments: 1. When I pull off the saran wrap, the sample sometimes comes off. The plastic in the sample turns white and dries out and does not attach. This is seen in the majority of my tissue sections. 2. The only time I have noticed consistent attachment is for a 5micron sized section. 3. How can I adjust the microtome so that the samples do not come off so scroll-like? My cutting angle is typically either 0 or 2.5 degrees. I also apply 70% EtOH to the block surface to soften the plastic. Overall goal: Get the tissue sections adhered to the glass slides so that I can proceed with staining. Thank you all. Hello Steven, I have used both of these antibodies for various projects and we performed all of these on the Bondmax from Leica with a 60 minute antibody incubation. We have used two FoxP3 antibodies from eBiosciences; cat# 14-4777 on human tissue and cat# 13-5773 for mouse tissue. (both were 1:100 dilutions and ER2 for 20 mins (EDTA pH9). I have only used Geminin on human tissue. It was from Proteintech Group cat# 10802-1-AP (1:1500 dilution using ER1 for 15 min-ER1=citrate pH6) All of these were on FFPE tissues for brightfield only. We did not use them for IF. If you need any other specifics, let me know. Jesse, I have discovered that there are many different ways to work with MMA. Probably no two techs are doing it exactly the same because there are so many steps in which to optimize it for your lab. From reading your protocol, I see similarities and differences. Rather than saran wrap, I use a thicker plastic that I cut into strips ( the size of the slide) from the plastic bags that we use for discarding paraffin waste. Although this is different from your protocol, I still think that what I do to remove the plastic might work for you: just before removing the plastic strip, I shoot it with a blast of freeze spray. The plastic just about falls off. Teri We do, it's more of a combination use log for the microtome and tissue flotation bath, we record temperature and that we cleaned the unit, used fresh water and also cleaned the microtome. This document also records the yearly calibration of the floatation bath and the yearly PM of the microtome. I do need to add that both the calibration and PM will have additional documentation that is stored in a designated binder. All Common checklist, COM.30750, Phase II Temperatures are checked and recorded each day of use for all temperature-dependent equipment and environments using a calibrated thermometer. I have seen these cassettes and while they are nice they are rather expensive and with re-embursemnt down everyone is looking at their budget. Also due to the design and the use of bar codes they do not always fit in the engravers. I still like the idea but with the amount of prostates we do my budget will be hit. Currently we use the sponges but have always had problems with getting the cores to be straight and getting a whole core in one tissue section. Plus counting the cores and correlating that at embedding. However, we are testing out a system called SmartBx that is an entire system starting at taking the Bx. It uses an imaging system to guide the bx, then a special bx needle system that places the core in a special cassette on a piece of treated paper - perfectly straight and flat. The cores adhere to the paper and the paper goes through to the final block using special molds at embedding. At grossing the bx/paper is simply placed in a cassette and sent to processing. At embedding the paper w/bx is placed tissue-down in the special mold and pressed down with an associated tamper. The final block has perfectly straight cores at the same level exactly 100um from the block face. There has been a learning curve but overall it makes prostate bx's very easy to handle in grossing and at embedding. the slides have full face cores on every single one. Laurie Colbert asks about processing prostate needle biopsy specimens. The subject is extensively reviewed in this month's or last month's Archives of Pathology. Your pathologists have copies of this if they're fellows of the College of American Pathologists. Here's the method as presented in that article - I've been doing this for a good many years now. The urologist should submit six containers, three for each side of the prostate, each container containing two or more cores. No more than two cores should be put in one cassette. Use those little blue sponges in the cassettes to keep the cores flat. Measure each core, and record the measurements. Don't dye the cores with eosin (fluorescent) - if you want to dye them use safranin O. Process with other small biopsy specimens. Embed using a tamper to get the cores flat in the bottom of the boat. Cut sections onto slides suitable for immunohistochemistry. Cut 5 sections of each block, with little if any trimming. Stain slides 1, 3, and 5 with H E. Reserve slides 2 4 for possible immunohistochemistry (needed in about a tenth of cases). It's a lot of work, but it makes cancer diagnoses. Expect a cancer diagnosis in about a third of cases.
