Hello histonetters,
Regarding my first email. The thickness of my section is 5 micron.

 I have a problem with my MMA plastic section on my slide. I could not remove 
the plastic MMA even if I leave the section slides in Xylene for several days 
with heat incubation at 60 degrees. This is the first time that happened to my 
section for several years of doing the same thing over. I was thinking that it 
was my Xylene lot number so I tried to change it but still does not remove the 
plastic, and then Itried different solvent Acetone, and Ethylene glycol 
Monoethyl ether but still it does not work. Can I ask for help if anyone knows 
what was going on and what would be the best way to remove this plastic from my 
section?

Just to give you my embedding solution was MMA -94% , dibutly phthalate- 5% and 
perkadox 16- 0.5%. This have been my solution for years and I do not have any 
problem with the removal of plastic section until now. I was thinking that my 
MMA (M55909) different lot number from Sigma Aldrich may have cause this 
because even I tried to use the MMA to dissolve my plastic it does not work.

 Hi,

We (at US Biomax) recently screened our colon TMAs for loss of expression
for MLH1, MSH2, MSH6, PMS2 using Ventana's antibodies.

If you need around 10 cases of negative tissue:
CO20813a, 208 cases of colon, contains loss of:
PMS2: 35
MLH1: 18
MSH6: 14
MSH2: 8

This TMA is not listed on our website but is similar in design to CO20813.
We have additional results on other 208 case TMAs too. Feel free to contact
me directly for more details.

Would anyone be willing to share what they use for control tissue for MMR 
testing?  Finding negative expressing tumor is becoming more difficult for us 
to find and we're wondering if there is an alternate negative tissue that would 
be acceptable.

NordiQC recommends using Tonsil as a positive tissue control and Colon 
Adenocarcinoma that has a loss of expression as a negative tissue control for 
all four antibodies (MLH1, MSH2, MSH6, PMS2).  Stromal cells in the negative 
tissue control should have positive nuclear staining and serve as a positive 
internal control.  NordiQC doesn't have any recommendations for alternative 
tissue to use as a negative control.

Anyone doing anything differently?
