technique. You could also do a working interview where a tech shows them around

[Histonet] Small processors

We are looking forward to ordering a new or used processor. We have the Sakura 
VIP5 however, I was wondering if there were smaller processors out there. We 
only do a max of about 60 skin specimens and the VIP is a bit large for that 
quantity. I would say a max of 100 specimens for future growth. Any thoughts.

We have 3 new leicas and we constantly have to adjust them. They creep forward 
so much that I just automatically know to roll it slightly back after every 4 
specimens or so. The new leica designs are just bad and customer service is 
even worse. We have to use the plate as we do Mohs specimens and the sections 
are just better with the glass plate. I want to look for a different machine 
next time. We did test the Avantik and it cut nice but the glass plate was on 
the front and got in the way. I wish they would talk to Mohs techs before they 
design these. Oh well. No tricks just constant adjusting. Thanks Anne

Hi everyone. Does anyone regularly use the anti-roll plate on their cryostat? 
Does it work well on the Leicas and Microms, or do you have to figit with it a 
lot? Does anyone have any tricks they can share to get it to work all the time? 
 I found that a little spritz of freeze spray on the plate and the bottom of 
the roll glass helps, but it is still seems a lot easier for us to use a brush 
or a teasing needle.

[Histonet] Stain line order

When doing frozen section staining for Mohs we have DI water and don't need a 
clearing agent. I am helping set up someone who is using tap water and needs to 
clear it. They mentioned that they used to clear first, rinse, then use bluing. 
I thought it was the opposite after heme. Rinse, bluing, rinse, clearing. I 
don't remember which is first. Actually do you need a bluing if using tap water 
since that blues better then DI water? Thanks for your input, it's been 10

[Histonet] Cryostats for Mohs specimens

We are shopping for a new cryostat; has anyone used the Tanner Scientific (r) 
TN50 cryostat? Can you let me know if you like it. We are unhappy with the new 
Leica's. We are also looking at the Avantik ones as well. Any input is helpful; 
this is for Mohs specimens so they need to cut well and be able to get a lot of 
tissue on each slide, sometimes the components of the machine get in the way of

[Histonet] Mart-1 rapid test for Mohs specimens

[Histonet] Frozen tissue

I'm testing a faster Mart 1 for melanoma on Mohs specimens. Its been a while 
since I've done any frozen tissue IHC stains and I can't remember the fixation 
to keep it viable for testing the next day, or same day but later on. Will it 
hold up if fixed in acetone and dried or do I need to refrigerate or freeze?

I have a back-up processor we keep filled for a back-up and if we have an extra 
run. However with lower volumes lately we aren't using it. I was just wondering 
about the paraffin in particularly. Does anyone know if you are not using it 
how long it can stay there (its heated) before changing out completely? Does it

[Histonet] Formaldehyde test kit question

We switched formalin neutralizing solution and I bought the Quantofix 
formaldehyde test kit to make sure it's working, however it does not mention 
what color is acceptable for neutralization. The options are 0-10-20- up to 200 
MG/L. should it be "0" for acceptable dumping? Also the instructions say to add 
10 drops of a sodium hydroxide solution to the solution being tested, wouldn't 
that change the Ph and make the results not as accurate. This kit is expensive 
if regular pH strips are better, let me know that too. Thanks for your help.

[Histonet] Xylene and Formalin monitoring

Just need a refresher on formalin and xylene monitoring regulations. Do we 
still have to do a 15 minute and 8 hour testing on both xylene and formalin, or

[Histonet] Grossing large ellipse melanomas

We are starting to keep our melanomas in-house now that we have a better 
processor, however I could use some tips or tricks on cutting large fatty skin 
ellipse specimens. My problem I think, is too small a knife, the specimens are 
not cutting even and the fat is getting in the way. So any suggestions or links 
to grossing techniques would be appreciated. Also, any ideas on better cutting

When disposing of slides after the 10 period is up, what is everyone doing to 
protect patient confidentiality? In the old days we would just put packing tape 
around the box and toss. With patient names on the slides and sometimes 
specimen location on the new labels I'm wondering how to dispose of them

If you don't already have it you may want to offer a shift differential for 
very early or late shifts and weekends.
This makes it a little easier to hire for and keep people on the harder shifts.

Good morning everyone.  I had a meeting recently with hospital administration 
where I was asked what leadership could do to retain and keep quality staff.  
Besides the obvious, pay increase's, I suggested a ladder type progression 
system for all lab staff, similar to what some hospitals offer nursing staff.  
My question to this group is, if any of you are using such a system would you 
mind sharing it with me?

[Histonet] Spill kits

[Histonet] Hoods

We had an inspection from the fire department recently and they said we need to 
have the exhaust from the fume hood checked annually.  Does anyone know who 
does that?  The people that do our P.M. on our machines say they don't, so who

You clearly don't know your histo history. The reason we know that H pylori 
exists is because a Scientist, Dr. Barry Marshall wanted to prove bacteria 
caused ulcers and not stress.  No one believed him.  So he took the organisms 
from a patient, mixed it in a broth and drank it. He then biopsied himself and 
treated it. There's a non-uniform method that saved a lot of suffering.  Bravo 
we crazy scientists.  Anne

Buy a bottle of what ever enzyme and use a reproducible buffer and 
temperature.

Yes, spitting is the tried and true way to do it.  Not to mention no measuring 
and cheaper.  The reason we switched to a powder is because I just don't spit 
well I used to have someone do it for me cause I would end up drooling. YUCK! 
The best way to find out is do the amylase method and the spit method at the 
same time and have the doctor pick the best.  A fun experiment  Anne

[Histonet] Height regulations
