Hello
Soak slides in xylene or equivalent solvent for sometimes till the
coverslip is loose, carefully remove the coverslip; you can remount the
slides with Permount.
You can dissolve the Permount in organic solvents, but the glue will be
dissolved and you have to remount the slide.

Hello
 Methanol is added to reagent alcohol (ethyl alcohol) so that people do not
use for drinking purpose and for tax purposes. Alcohol (ethyl alcohol) has
federal tax just like liquor.
One can buy pure alcohol, but you have keep inventory and make sure it is
not misused.

I have been reading about xylene, how dangerous it is.
Have you thought about using mounting medium made from Limonene.
Here is some info:

*AR-6504 Organo (Limonene) mount™:** *This mounting medium is made with
limonene a natural product from orange peels. It is good for preserving
tissues and cell smears that can be dehydrated with organic solvents in
Immunohistochemistry (IHC) e.g. DAB and DAB with nickel or cobalt. This
Organo mounting medium is also suitable with alkaline phosphatase
chromogens, an organic solvent resistant Super Fast Red (IBSC, cat AR-8211).
It is an excellent choice for mounting H and E stained slides. Coverslip is
required. *DIRECT SUBSTITUTE OF Biomeda’s Clarion*

[Histonet] Light + IF microscope

I need your professional advice.
We are a small IHC and IF company looking for a good microscope.
We have investigated in Nikon;s they have 50i and 55i microscope that one
can use for IHC and IF with proper attachment, but pretty steep price.
Do you have any recommendation for a good quality microscope w/ IF
attachments.
May be one of you have a microscope like this and want to sell at a
reasonable price.

[Histonet] Mounting medium for IF

Pi gives red color in IF, it stains DNA and RNA

PI excites at 535nmand emits at 615nm, producing a *red* fluorescence.

Therefore if you are using any chromophore at these wave lengths which emits
red color you will have problems.

By the way we also have several mounting mediums for IHC and IF, if you
would like to try, please let us know.

I have the same experience.
When I worked for a company and right after 9/11, we shipped antibodies to
Israel via Post office at ambient temp.
This package came back after 6-8 weeks or may be longer.
We do not know how they were stored at post office.
We tested the antibodies and could not find differences in the activity
using IHC.

[Histonet] Plants in Histology lab

Everybody is writing about the plants in the histology lab.
May be I will share my experience with you in Biochemistry lab at MSU .
Many many years ago when I was a post doc in Biochemistry dept. I had lots
of plants in the window.
One day when I came from lunch, I saw unusual plants along with mine, on
close look, they were marijuana plants.
I almost had heart attack to see these plants in my cubicle, right away I
hid these plants.
On investigation we found that our dish washer had these plants some place
in the lab. and she was hiding it from the professor.

I need some help and advice from you experts.
We are new in cutting formalin fixed paraffin embedded (FFPE) tissue
sections.
We need your advice.
We have a used Leitz 1512 microtome.
--- perhaps it needs service, can you recommend a good and reasonable
place.
what  will be a good disposable blades to buy for cutting FFPE
tissues sections (mostly soft tissues like colon, stomach, skin, cancer
tissues  etc).
this  instrument needs a blade holder, what would you recommend,
Leitz does not sell blade holders for this instrument anymore.
any good technique for cutting tissues for a person who is not
expert.
good place to buy formalin fixed tissue blocks for research use.

Hello
My experience is that most Immunofluorescence dyes do not withstand alcohol
and organic solvents dehydration.
You can use aqueous mounting medium with anti-fading agent.
Frozen brain tissue is tough to work with;  since it contains tons of
lipids.
Lots of mounting mediums will produce air bubbles because aqueous mounting
medium being hydrophilic and brain tissue being hydrophobic.
We are in the process of developing an aqueous mounting medium with
anti-fading agent and should be OK to use with tissue like brain, if
interested please let us know we can send you a sample for evaluation.
