If you could share the responses you get I'd appreciate it.  
We just built a new facility.  There are cameras everywhere.  They are in the 
lab and I hate them but you soon forget that they are there.  They cover almost 
all of the lab.  There is a small area that isn't covered and we call it the 
personal scratching area.  LOL .  My office does not have a camera in it.

We have put eosin in the last alcohol on the processor for years.  It stays in 
the esophageal biopsies just fine.

reguarding the eosin biopsy post on the histo net. Things you might want to 
considered looking at, I cannot say for sure because I don't know your 
processing solutions/schedule:

Most Eosins are alcohol soluable, by putting the eosin in your formalin most of 
it will wash out during processing. Many labs put the Eosin in the last alcohol.

Hematoxylin is typically water soluable, if you put it in formailin processing 
usually removes water if the formailin is followed by alcohols.

So, I work in a small GI lab, and I put Eosin in my first formalin on my 
processor.  My biopsies are very small and this helps, somewhat, to see the 
specimens for embedding and cutting.  But, unfortunately, the esophagus tissues 
do not absorb the eosin much.  Anyway, the hospital lab I work, part-time, in 
has started using hematoxylin to help see their biopsies.  I happen to embed 
there and I think it just makes a big mess and the tissue does not absorb much 
of the stain.
What are other labs doing to aid in making their small biopsies easier to see?  
What are pros and cons to doing this, in your opinion?

Now you're making me think!  I do have a cut-off.  It's at 95%.  I do keep 
records of the percentages of specimens that do not fall into the range.  But 
as to the process of solving the problem it's just counseling.  As we have only 
hit that cut-off once in the past three years I really haven't thought it out 
farther than that.

that's exactly what we do here.
I wondered, if you fixed a cut-off. Because in quality-audits they often
asked the "what do you if.."-questions. 
for example: "There is an increase in the number of discrepancies in this
special month. The rate is over your defined aim. What did you do to fix the
problem? Do you keep records about the process of solving the problem?"

I was just curious, if you go further than the counting of events. 
In daily work we often know the causes of mistakes, and very often we know,
that a certain amount of mistakes can't be avoided, although everyone tries
to give his/her best. 
I think QM should help to discover the special events or circumstances, that
raise the number of mistakes above the "normal" level. I was curious, if you
defined this "normal" level for your lab.

gross is written on the side of the cassette so that at the time of
embedding the tech knows what should be present.  That helps in being sure
there isn't a piece stuck to the lid or forceps, or popped out when opening
the cassette.  It's really more of a Quality Assurance measure than tracking
a discrepancy.  I do watch how frequent there is a discrepancy though and if
there seems to be a pattern will initiate a review of processes for both the
grossing person and the embedding person.  
What we see most is the gross will say one piece and there will actually be
multiple tiny pieces or two pieces will be three at embedding.  As all staff
is aware that we are following this they tend to be much more precise in
counting at grossing and embedding.

Quality Issue and just a deeper section.  If a section is folded, has chatter 
or some other quality issue then I think it needs addressed as a Quality

b.  Frozen section vs Final diagnosis correlation

I liked the idea of piece count discrepancy or even poorly processed blocks.  I 
never recommend using "recuts" as a QM because that is more often a product of 
the pathologists' preference or desire to see more tissue and does not reflect 
the quality of the sections (unless it is an incomplete facing issue).  This 
can quickly become punitive for your department because this information is not 
presented to people who understand pathology.

We print an average of 150 blocks a day and 250+ slides a day.  We have 2 
cassette printers and 1 slide printer.  We have had them for several years now. 
 They are Primera printers purchased from Creative Waste Solutions.

Also they can connect to your LIS if you want very easily without needing an 
expensive interface.

I've used the Primera slide printer since it came out.  I like it a lot.  It 
needs it's maintenance kept up like anything else but other than cleaning the 
rollers there is very little else that needs cleaning.

I totally agree with your assessment of the Slidemate printers. Have not had 
any experience with the newer model. 
Applying slide labels shouldn’t be an issue when dealing with one block and 
slide(s) at a time.  Some of this could depend upon your software available to 
you. Our system generated just the labels needed for the block you were 
cutting, on demand. You should be able to get a nice Zebra printer for under 
$700.

Another of our labs used the new Sakura/Premera printer and really like it.

We have used the older version Slidemate slide labelers from Thermofisher for 
the last three years.   I think putting the patient information directly on the 
slide is preferred however the wear and tear on slide printers means higher 
maintenance costs and more down time.  Thermofisher has a newer version of the 
Slidemate now that has been designed so that there is less wear and tear on the 
machines.   Currently when one of the old labelers breaks down we have to send 
the instrument out and the cost is generally 1000 - 1500 for repair each time.  
  It also seems that the older machines have to be sent in at least every other 
year.  Our older machines have been sent in almost yearly.

We are considering switching to a slide label making system by General Data.   
One of the downsides is that we have to apply the label to each slide so 
therefore we introduce another step where the slides could be mislabeled.   The 
upside is that equipment costs are much less.   A new thermal label printer 
runs around 700.00 where as the instrument that imprints the slide label on the 
slide runs between 12000 - 15000 per unit.  We need three units.
Thermo also offers a lease system.What I need to know is what has been the 
experience with the new Slidemate AS printers?   What kind of maintenance 
issues have you encountered? Finally please let me know your latest experiences 
with both slide label printers versus slide labeling printers?

I really take exception to those finding fault with this book!  Maybe another 
word other than hack would have made some people feel better but we all know 
the intent.  Stating that "many of the fixes or secrets described are because

totally incorrect.  Very few of the "hacks" has anything to do with someone 
failing to do their job.  I think this books intent was to help new people in 
the field of histology.  It has been a great form of dialog between our new 
tech and the older techs.  It has served a very good purpose.

Kind of in that same vein, but not to denigrate the book ( I have not seen it), 
I try to impress on people that "enabling" a poor process via "workarounds" or 
"shortcuts" does nothing to solve the original problem - it just prolongs the 
problem and peoples frustration with the problem. A much better path is to 
figure out why people feel the need for a workaround and then solve that 
problem. In other words, if a workaround is needed, then you have a problem 
that needs solving!

So now I have people come to me  saying  "I think this thing we do is a 
workaround for another problem." That is the kind of observation you like to 
hear.

I purchased the book and applaud the effort because there is some decent 
information.  However, the term hack is a poor choice for histology and many of 
the fixes or secrets described are because some hack failed to do her/his job 
at an earlier step in the process. (Definition of hack. transitive verb. 1 a : 
to cut or sever with repeated irregular or unskillful blows. b : to cut or 
shape by or as if by crude or ruthless strokes. As a noun it is used to mean a 
mediocre performer or worker; tiresome drudge.)

Some methods, while useful in some settings, have important cons not listed, 
cons which may be counterproductive. For example using Mercurochrome or Eosin 
to mark tissue may preclude further testing with fluorescent endpoints, such as 
FISH.  Plus if you really want to use Eosin to mark the dermis, it is far 
easier to add used Eosin to one alcohol in the tissue processor. That gives a 
visible indicator of carryover, indicating need to change or rotate solutions.

Other methods seem (to me) like workarounds or Band-Aids for Labs w poor 
grossing, poor processing or poor reagents or poor technique or poor method 
choices, eg, 2.14 describes a situation where an incompetent grosser truly 
hacks or crudely cuts into unfixed tissue yielding too thick a slice. The real 
solution is to fix the tissue before slicing. Poor fixation results in poor 
processing and poor sectioning and poor staining reactions.

For another example, Cassette sponges offer few advantages, while folding lens 
paper allows the grossers to see through the paper and know all pieces are 
inside before closing the cassette lid.  The tissue does not stick to it, and 
small flakes can be scraped from the lens paper at embedding. Last during 
folding, the forceps can be cleaned at grossing and during unfolding, forceps 
may be cleaned w the paper after embedding.  Sponges also result in greater 
solution carryover.

Several colloquial naming conventions, eg, chamber saver 2.16 for 
underprocessed tissue may be memorable to some readers, yet seem  are odd to me.

This 2.16 method is an especially useful technique which may also be done to 
extend paraffin time, whenever poor sectioning due to poor processing is 
encountered at the microtome.
Variation 1 Place the block back into the proper sized mold and return to the 
heated side of the embedding center for an hour to extend processing 
(reprocessing). Then remove the old paraffin from the mold w a plastic pipette, 
 then re-embed, replacing the paraffin w new.
Variation 2 for outside blocks we routinely replace an unknown paraffin from 
another lab by melting in a mold, and 'reprocess' in our (blue ribbon) paraffin 
for 1 hr in a mold in the embedding center then re-embed.

Good first effort, yet this book could be improved by a good editor, by more 
collaborators, by illustrations, and the addition of variations or other uses 
as described above for 2.16.

book has some things we had forgotten too!  We all have been enjoying reading 
and adding little comments.  Since we are a GI lab it was great to have some of 
the other hints that our new tech hasn't had much exposure to.

Would any one advertise in the local paper that your center of excellence lab 
loses nearly 1% of your specimens?

prophesy, one to be eschewed and denounced, or beaten out of the grosser who 
dares use that description for it is an expectation of failure.
That grosser would not last a week around me.
I would walk them to the train station and ask them not to return.

I would think that it would really depend on if it is a repeat due to an 
instrument problem or a human error.  I think that if a vendor says that a 5% 
repeat rate is acceptable for their instrument I would look for another vendor.

Rachel,

We dewax off line.  We use the same protocol that we do for any other staining; 
3 min in  3 changes of xylene (we use a xylene substitute though), 3 min in 2 
changes of 100% alcohol and one 3 min in 95%.  
We use BioCare's IntelliPath immuno stainer.  It is a completely open system.  
All of the sales reps, support techs, and service tech have always been very 
open and willing to help in any way to provide information and support for this 
stainer.  If by dewaxing you are also referring to off line antigen retrieval, 
we use the pressure cooker that BioCare has.  You can use any retrieval 
solution you choose with it.  If you need to use two different retrieval 
solutions you can fill your containers with two different retrieval reagents.  
I don't find that there is any difference in doing off line retrieval than on 
line retrieval.

Does  manual dewaxing affect the staining? Every rep says yes... but does it?

What is the time for dewaxing when you start the instrument? What are the 
number of steps in the protocol? What is the waste generation like more or less 
than manual?

I have run into quite a problem.  My lab currently processes all tissue types 
from 3 different sites.  Recently, we have been getting complaints from one of 
the sites that the biopsies have a nuclear "artifact".  It is described as 
"washed out" or poor to no nuclear detail.  Pictures have been uploaded 
(Nuclear "Artifact").  The Medical Director at said site is convinced that a 
processor error is occurring.  Our site is not seeing this on any of our 
slides.  Biopsies from all three sites are processed and embed together.  We 
have done all trouble shooting that we can think of.  Leica service has come to 
inspect our Peloris processor and all areas checked out as functioning 
properly.  The problem is not consistent daily.  Seems to be worst toward the 
end of the week.
We have been running the same processing protocol, staining protocol and 
cutting protocols for years now.  This problem has just developed over the last 
2 months.  Any ideas, no matter how far-fetched, would be greatly appreciated 
at this point.

Fortunately times have changed.  The person I replaced in the late 70's early 
80's had been brought in from the hospital laundry and was "trained".  They 
were still pouring embedding molds then.  She did a beautiful job at cutting 
and staining H&E slides and 2 or 3 specials but that was all there was to do.  
This world has come a lot farther than those days of the 80's and 70's.  We 
have grown into fully capable labs that don't have to send work out to the big 
reference centers to have tests done.  I remember a class with Lee Luna when he 
said "We have to excel and learn these immuno procedures or the MT's were going 
to take them away from us."  We learned and they didn't!  We may not be 
recognized at the level with MT's but we are slowly changing and getting there. 
 At least now an associates is required and CEU's are required.  I think that's 
progress.  If you're looked down on in your present position move on!  It's not 
worth it.

This is a fight that we continue to have with hospital administration.  In my 
opinion histologists are just as important and needed as MT.  Even though there 
is an increase in automation in pathology the hands on of a histologists is 
most important.  The fact that hospital still consider a lower entry job is the 
reason there are not more of us.  It is quite frustrating.

Tanya, you can probably put either date/time into the receive date/time prompt 
of your LIS.  It just depends on how you want to use that information.  For 
instance, perhaps you want to look at turn around times.

is that those cases received off shift are going to skew your turnaround times 
if that's something you measure.

Bottom line, it just depends on what you're using that information for.

I am curious as to how everyone handles the receipt of their surgical, and 
Cytology specimens in to the lab? Especially on the off shift hours? Other 
clinical lab specimens are received directly in the computer in Specimen 
Processing when they come in the door from remote locations or via the tube 
system, then delivered to departments. During the day, specimens get dropped 
off directly to Pathology by the various departments collecting (ie. OR, 
Radiology, etc). On our off shifts, specimens get received by Specimen 
Processing(date/time stamped and initialed)  and put in the fridge for 
Histo/Cyto the next day. When Histo and Cyto put the specimens in the computer, 
for the "receive date", they go by the date the specimens are "received" (or 
unpacked) in Histo/Cyto. I am thinking we should go by the date/time Specimen 
Processing received them in to the actually laboratory where the specimens are 
then "controlled".

I also have a Primera slide printer. It worked well at first but now it is

Hi all!

When I took HTL exam and there was a question on the exam that bothered me the 
most but I cannot find an exact answer for it, had to guess!  I'm very curious 
about your thoughts on this.  I have asked Histology Core and they weren't sure 
about it either - wanted to share it with you guys if you know the official 
answer for this!

The question was mainly about the angle of wrist/hands on the rotating 
microtome and the degree must be correct so it won't cause any stress on 
wrist/hands, it listed 4 different choices:


1.   30 degrees wrist down forward

2.   90 degrees straight without bending the wrist

3.   60 degrees wrist up backward

4.   60 or 90 degrees wrist down forward


(Please be gentle with me since I don't remember the EXACT numbers but those 
were close to numbers provided.)

How about the person in the 1970's coverslipping with an open dish of xylene at 
AFIP and someone at the other end of the stain line decolorizing Brown and 
Brenn stains with acetone/ether in the sink; then the acetone/ether fumes

and wall catch fire.

    Ahh, the good old days of walking into the lab and lighting all the Bunsen 
burners first thing in the morning.  I didn't have any hair on my knuckles for 
years.  Embedding with a Coke on the cold plate, and a smoke in the ashtray 
next to you, anyone?  Good times.

    Now we have to use forceps warmers and change forceps between specimens.  
If you get 3 or 4 pair of forceps, one will always be hot enough to use.  Also, 
there are embedding centers with heated forceps, which I love .
    Just remember to clean out the wells of the forceps warmers every day to 
prevent cross contamination.  Cotton applicator swabs work great for this.  And 
always keep a towel or gauze handy to wipe the tips of the forceps between 
specimens.  Forceps warmers unfortunately don't incinerate any stray tissue 
like a Bunsen burner did.

You might find that the financial impact wouldn't be that great.  There is a 
reason they are doing an EGD, most commonly for gastritis which is a valid 
reason to look for H pylori.  There is a pretty standard list of ICD 9 codes 
that would trigger the justification for doing an H pylori.   Also, you're 
going to one of two things when you have a stomach biopsy.  You will either cut 
extra slides, (time and material there) or you will wait until its ordered 
(loss of TAT).  So, if you are going to do a retrospective study you need to 
include that in your review.

You might be able to get around this by doing the IHC on all of these as 
requested, but only charging for those which are indicated as being medically 
necessary. This could benefit your TAT, if that's the reason the pathologist 
wants these done routinely. The pathologist would also have to agree not to 
bill for the PC of this test in these cases also. You might want to do a 
retrospective study and determine if this was done what the financial impact 
would have been.

My understanding is CMS (Medicare) has ruled that h.Pylori stains cannot be a 
standing order.  The Pathologist must look at the H&E's first and deem the 
stain is needed based on the inflammation.

I understand the 88343 codes to be for multiple stains on the same slide. The 
first is 88342 additional 88343 - e.g. PIN4.

Actually, for non-Medicare if there are multiple stains performed on a block, 
where the initial CPT is 88342, any subsequent billing for stains on that block 
would actually be 88343 from the information I see.

additional single or multiplex antibody stain (List separately in addition to 
code for primary procedure)) to ensure that the services are only reported once 
for each antibody per specimen. We believe this will result in appropriate 
values for these services without creating incentives for overutilization."

clarify what this all means.

I agree with Tom.  With the exception of self-inflicted issues we also have not 
had any issues with recycling our reagents.  We check each batch as it is 
recycled.  
We also don't have a problem with fumes.  (And our pathologists are fussy)

While I can agree that recycling alcohol and xylene is both environmentally and 
economically advantageous, technically it's awful.  There's no way to make used 
alcohol and xylene as pure as it was originally.  There's also the issue of 
fumes from recycling a solvent.  The company I had used years ago swore that 
there were no fumes when using their machine but the personnel working in the 
laboratory would vigorously disagree.

The life of a basement lab rat isn't all bad!  We don't hear thunder storms.  
We are probably be safe from tornados, hurricanes and will have lots of company 
if one does come by.  Usually no one wants to go to the basement.  
There are no windows so there isn't any of the outside world distraction.  If 
it's a beautiful day you don't see it so you don't long to be out there.

by windows once in a while. :)  But for some reason we do get paid more than

We use Formula 83 from CBG for both processing and staining.  We have used this 
for years with no problem for our GI biopsies.  It wouldn't do any good to

Instead of xylene you can use isopropyl (also known as 2-propanol and propyl 
alcohol) and after sectioning you can use dishwasher soap before going directly 
to the staining procedure. After staining you need nothing but an oven to dry 
out the stained sections and go diractly to coverslipping.

Here is what I have in my notes for the new standards.  I think you're probably 
safe with what you currently have as long as you've don't  the training that 
should have been done by Dec 1st 2013.

The first compliance date of the revised HCS is December 1, 2013. By that time 
employers must have trained their workers on the new label elements and the SDS 
format.

All hazardous chemicals shipped after June 1, 2015, must be labeled with 
specified elements including pictograms, signal words and hazard and 
precautionary statements. However, manufacturers, importers, and distributors 
may start using the new labeling system in the revised HCS before the June 1, 
2015 effective date if they so choose. Until the June 1, 2015 effective date, 
manufacturers, importers and distributors may maintain compliance with the 
requirements of HazCom 1994 or the revised standard. Distributors may continue 
to ship containers labeled by manufacturers or importers (but not by the 
distributor themselves) in compliance with the HazCom 1994 until December 1, 
2015.

 We use Leica's Selectech also and occasionally have the same issue.  I don't 
think your ETOH after the eosin is a problem.  If you add a 95% you will leach 
out the eosin a bit.  I think you should add another 15 - 30 seconds in the 
acid alcohol.  One of the things we find here though is that there can be a 
great variation to the pH of the water.  That affects the results of the stain. 
Specifically the "purple haze"  Go Jimi!

Hi all-
Our histopathology lab is looking to
set up an IHC component to service the request of our clients.  All work is on 
animal tissue.  We will probably run about 1,000 IHC slides the first year and 
hope to increase the workload each year.  Some  stains we would run would 
include CD4, CD8, Ki-67 and TUNEL. We looked at the Ventana Discovery XT and 
appeared to be a good fit for our needs.  Does anyone have any other advice on 
other machines we should look into?  Any information or advice would be greatly 
appreciated. Thank you!

That's what I was going to suggest.  "open a drawer and put a tray or board on 
it"  We do that any way as a place to set the slides and other paraphernalia 
that you may need.  It keeps from having to reach over or around.

I would suggest that if you have a table (or can open a drawer and put a tray 
or board on it), put your water bath beside you. Then you wouldn't need to 
"push off". Maybe that would be a solution?

Thanks Peggy,

  I've heard many positive accounts and they help me maintain a positive 
attitude.  My main restrictions, for a few months are; no crossing of legs, no 
turning of feet or knees inward, and no bending at the hip past 90 degrees.  
Also, items need to be located between my legs before lifting.  One of my 
concerns is the ability to move between microtome and water bath, while 
sitting, without violating those restrictions.  I may need to get creative in 
the layout of the lab.
 On a positive note, the Leica RM2255 we purchased earlier in the year, 
helped alleviate my repetitive motion problems.  Thank you Histonetters for 
helping me with that selection.  I apologize for being so long winded.  I guess 
spending the days locked inside, interacting only with a couple snoring 
dachshunds, might be taking it's toll.

We use to use a paraffin floor scraper kind of like a scraper on the end of a 
broom stick but somehow it disappeared.  We now use a paraffin scrapper that is 
much easier on the floor.  The one designed for scrapping the floor had a blade 
that had a tendency to gouge the floor.

1.  Making formalin and Grossing with an old metal table fan to blow the 
formalin fumes away.  
2.  Finding the technicon hung up in the morning with the samples dried out.
3.  Figuring out how to reprocess the tissue
4.  Pan embedding and having the paper labels float away from the samples 
in the pan.
a.  Bunsen burner needed
5.  Big kitchen knife heated over a Bunsen burner to make blocks
6.  Soaking re-useable metal cassettes in a bucket of xylene.  (next to 
where you were pan embedding)
7.  Sharpening a knife on multiple stones then stropping on leather then 
cloth.
8.  Over tightening the microtome clamp on the paraffin block and having to 
re-embed because the block broke.
9.  Making staining solutions (some of us still do this).
a.  Making Schiffs regent without a hood
b.  Cresyl violet stain powder floating around the room,  watch 
where you put your hands
c.  Silver nitrate spilled on the counter where you just put your 
hand then wiped your face. 
10. Staining slides in open staining dishes with no fume hood.
11. Wiping the whole room down with xylene to clean the wax up.
12. Autopsies in the afternoon,
13. Whole human brains soaking in the sink in 5 gallons of 20% formalin 
overnight.

Oh boy,  really miss them good old days.  Life was easier then!

Hey Teri,   When are we going to get our new coverslipper in?  We were 
spoiled by the demo unit.

I've stropped blades until they looked like scythes - and I got pretty good at

Seems like no one has touched upon the fine art of knife sharpening. Sometimes 
I felt like an executioner sharpening my axe to the point of splitting a hair. 
Heaven forbid if you hit a staple. That part of the blade was no useless for 
days to weeks until the daily sharpening ground it out. Life is pretty good 
when you just pull out another blade from the plastic box.

uphillboth ways. . We had to stock our cryostat with fresh cut blocks of 
ice from the river 10 miles away. (uphill, both ways) We had to start our

The issue remains the WE cannot impose on others how we are viewed. No matter 
how many brain storming meetings there are about "demanding" respect the issue 
remains that those who have to respect us have to do so.
Short of a national strike (absolutely impossible to achieve) or a national 
union (that will be fought to the death) the only thing we can do is to stand 
for our rights of being treated as professionals every time somebody, either 
pathologist or administrator, disrespects us. And who, may I ask, will be able 
to do that in an economic situation as the present one? Who is going to risk 
his or her job just "to demand respect"?
That is an illusion. Unfortunately the economic situation impedes any action 
and those who can will keep disrespecting us as long as they can get away with 
it.
Having a good "position statement" about "what does it mean to be a 
professional" is an exercise in futility that probably for those who wrote it 
may have meant being able to have 1 or 2 days paid without actual work to do.

Hi,
I understand using lean at the embedding center but when it comes to the little 
things like the types of forceps it may be best to let the techs use what works 
best for them.  Everyone's hands are different and the feel of the forceps in 
their hands and dexterity come into play here.  If you have many different 
embedders maybe it would be best if each had and were responsible for their own 
forceps.  I don't know how that would counter affect the lean process.

What is the best way to label blocks that are already embedded?  We have a 
large control bank that we are trying to relabel and make search able, however, 
most marking pens don't write well on paraffin covered blocks, even when we 
scrape them off.  Pencils work, not always the most legibly, so I was wondering 
if there is a clever solution that someone might have found.  We've tried 
putting stickers or labels on the back, but that falls right off.

I don't think it's a matter of being reasonable or not, though I think 26 
blocks is a bit high.  
The major issue is taking away the discretion of the tech to make a decision as 
to the quality of the slide they are producing.

66 blocks X 20 workdays a month = 1320 blocks a month

Kiran,
We use Formula 83 on both staining and processing without any problem at all.  
You have to watch and either rotate or change your last Formula 83s on the 
stain line because you can't tell if there is water in it like you can with 
xylene.  We also use CBG's recycler to recycle Formula 83 without any problem.  
We have not had any problems with our IHC using Formula 83 either.

Bea,
You can use need to use the clipped corner slides.  The best ones are the Epic 
Scientific slides available from CWS, Inc or the clipped corner Tanner slides 
available from Mercedes Medical.  I'm sure others carry them also.  These are 
the only two slides I've found that work really well in the printer.  I am very 
happy with my slide printer. Set up was very simple and interfacing with our 
LIS was very easy.

I have the Primera Signature Slide printer made by Primera Technology and sold 
by Creative Waste Solutions.  http://cwsincorp.com/
I really like it.  You can use it as a stand-alone or connected to your APLIS.  
It's compact and works very well.  You can print your slides with a color band 
across the top to distinguish different stains or types of tissue if you want 
or add bar coding.  I don't use bar coding yet so I can't say how well that 
function works.  The printing on the slides is very good.  There hasn't been 
any smudging or fading. There is a little bit of babysitting but not much.  
Tech support is fast and reliable when needed.

We have used the Decloaking Chamber for several years now and have had 
excellent results. The nicest thing is that there is consistency and 
reliability.  Each run will be the same as the one previous and having the 
ability to use the USB stick for documentation of time, pressure and 
temperature makes record keeping really easy.  We have had no complaints.  The 
only con I can come up with is that it comes in a big box that is hard to store 
if you have to send it back for some reason.

with our current method using a vegetable steamer. We use many different tissue 
types both animal and human. Also we use many different antibodies. Currently 
with the vegetable steamer we've used Citrate, EDTA, and Tris-EDTA 
combinations. We've used times from 30-60 minutes, and even tried 30 minutes x 
2 or 40 minutes x 2, but we still see some inconsistent staining. To make sure 
it wasn't due to fixation vimentin staining was performed and shows even 
staining. It's just certain antibodies that are giving us trouble even from 
multiple different companies. Any advice would be greatly appreciated and very 
beneficial to our work pertaining to the instrument.

We have a check sheet that has all cases scheduled for the day.  There is a 
place to log how many specimens were collected for each case.  If there were no 
specimens collected then that is logged also.

I have used tap water without any problems.  The only thing is you need to be 
sure you clean your waterbath well at the end of the day and dry it out.  What 
I experienced was hard water residue on the waterbath.

I feel like I am racking up frequent flier miles on Histonet lately:) We are 
seeing bacteria in our type I.  This requires us to obtain our Type I from 
another of our lab sites.  Do I have any other options?  Like tap water?

We have used Formula 83 from CBG for years with excellent results.  We use it 
for processing and staining.  We also recycle it making the cost very low.

I know this is out of the box, but it happened to me so I thought I'd pass this 
along. We had the same issue with our chief pathologist...hematoxylin was 
too light. We tried everything, but she still complained!! Then we diluted our 
Eosin Y 1:1 with 95% alcohol and then she was happy. Crazy, I know, but 
something to think about!

I am having a problem with my staining, ( well, I think its pretty good but the 
path says its not cark enough for her) So, I am using Azer Scientific 
Hematoxylin Extra and I initially started out with 2.5 min in it. I upped it to 
5  min and then to 10 and 12 min. The problem is the control I have works and 
looks great at the 2.5 min. Besides just keeping it in the heme longer what 
else can I do? I have cut a few extra dummy slides today so I can try different 
things out with them. I am tired of hearing from the path everyday that the 
heme is too light for her.
I am a new lab for a derm clinic and I work alone so its basiclly up to me to 
figure this out. Please help! Thanks!
