[Histonet] Farewell
Hard as it is to believe - and I probably won't until I've been away for a week or more - this is my LAST day to work. Retirement? Who knew it would come around so fast? I remember having only fifteen years left to work and that was just yesterday! Histology has been 'bery, 'bery good to me and although I fell into it by accident, it has been a fascinating, involving, liberating experience. I'll be lurking on Histonet but under the alias of nmhisto and I'll probably do some p.r.n. work but I'm going home and throwing away my alarm clock and resetting the coffee maker for 6:00 a.m. instead of 3:45 a.m. Thank you all for your advice, guidance, humor, relative insanity and wisdom. Enjoy what you do and try to recruit at least one person into this field before it's your time to pick out the color of the tennis balls on your walker. [Histonet] Control Block Tracking
During a recent inspection of my department, the inspector encouraged me to develop a method of tracking which control tissue I used for a case (i.e., individually identifying the block and referencing that identifier on the slide). To be honest, I believe this to be overload, but since I'm probably going to have to abide by that encouragement, I'd like to know if/how you identify which control block is used for a special stain. I can think of a simple method but wonder if any of you have used a really spectacular method that just blew the inspecting agency away. I'd promptly plagiarize it! Gracias! Thank you to everyone for your tracking suggestions. I think we all agree that there are reasonable regulations and requirements for QA, and we can also agree that some are doing nothing but making our jobs more complex when it's really unnecessary. Soon, every laboratory professional will be attached to a tech robot - your clone physically! Every time you make a move, the docutech photographs and documents. At the end of the work day, you unplug your docutech and put him/her/it in your locker for recharging. I'm thinkin' there must be a movie in here somewhere - and it may not be a sci-fi feature. [Histonet] Removing coverslipping tape
Soak the slide in acetone for 10-15 minutes; it turns to something that looks like a slice of Jell-O and slithers down the slide, leaving cells and stain and structure in perfect order. I usually dip the slide in xylene a couple times to clear the acetone and re-coverslip it. Works like a charm. LOVE the coverslipping tape! I'm down to 20 days until I retire! Not that I'm counting or anything... I don't suppose that anyone out in HistoPersonLand has done a cost comparison between making your own 10% NBF and purchasing a 50-gallon drum of 10% NBF, perhaps? I did this several years ago but my idea of saving tech time/costs by buying the bulk was shot down because there was someone here that did the making. Now we're even more shorthanded and I think it would be a better use of everyone's time to just buy the darned stuff in bulk. Any chances that someone's already done this? Although I appreciate individual opinions, do not overlook the Leica line. Us old timers may recognize the names Sakura or Thermo (in its many permutations) more readily, I'd put my money on Leica every time (and I have - with processor, embedding center, microtome and autostainer). They do not let me down. I have no issues with either other vendor, but I placed my trust in German engineering. AHA! I have an Answer! EMS (Electron Microscopy Sciences) sells a One-Piece Scalpel Blade with Handle Cat. #72048-01 that is perfect for cleaning blocks of excess paraffin! I've had one for over 30 years and it will retire with me! It is initially sharp (but not as sharp as a normal blade would be) but if you cut open a few cardboard boxes with it (which it also does beautifully), it dulls down to the perfect edge for cleaning blocks. It fits the hand well and doesn't shave the plastic off the cassette. [Histonet] Lab humidity range
We're prepping for a Big Inspection (I could tell you who but then I'd have to enter the Witness Protection Program). The Inspecting Organization will want me to cite a range for HUMIDITY in the lab. Because this not only hinges on our weather (we're in Monsoon Season right now and the lab humidity is 47% as I write but is normally between 17-20%), but also MUST be based on any of the histology equipment that may be affected by humidity in any way. Realizing that we all work in different environments and usually have little or no control over the HVAC system, what did you decide upon as your HUMIDITY RANGE for the SOP under which you operate? If there are no specific equipment requirements, I will be able to give a range of 0-100% without the necessity for a Corrective Action Report. If I am too specific and give too narrow a range, I may end up filling out a Corrective Action Report for days when I'm out of range. Please give me as much detail as you are able, without being required to join me in hiding. Your input will be rewarded (don't ask). Do you use a remote alarm on your processor? If so, would you tell me what brand/model/manufacturer and how it works (keeping in mind that I am Electronically Challenged)? [Histonet] OT: Wednesday Half-Wit
I used to use Sub-X in the processor. Changing paraffin was no problem because I'd just drain it into a Styrofoam box, allow it to set, seal it up, mark it waste paraffin and put it into the regular trash. Easy, right? Well - then I changed back to using xylene. First time I changed paraffin, I used my tried-and-true method, only to arrive at the lab in the morning to find a partially dissolved Styrofoam box on the counter, surrounded by a solidified pool of paraffin. Eureka! Xylene dissolves Styrofoam! It was not pretty but I think I've invented a new space-age compound. Fortunately, I was struck by an Inspiration - use an empty xylene/alcohol bottle with the top cut off (so I can insert the paraffin drain). I don't know why I'm posting this except to show that when confronted with an unusual circumstance, women rule! Now, back to work. [Histonet] New Methylene Blue
Okay - it's not for me, but I have offered to make up a new solution of NEW METHYLENE BLUE for our specimen prep area. I need a recipe for making an aqueous solution used to stain fluid preps from animals (urines, etc.). I've checked Lillie, Carson and Sheehan and Google. Can't seem to find an appropriate Formula. Help? [Histonet] Retirement
It seems that my reference to RETIREMENT has gotten everyone thinking about it. Heh...heh.. It has been suggested that I reconnoiter in advance of Those of You Who Won't Be Retiring Before I Do (February 29, 2012, if the creek don't rise...). I would be happy to perform that hazardous duty but I need more of those $5.00 donations coming in for whatever it was that I posted last week (I hope my gray cells will rejuvenate when I retire). I won't need the money for travel because I think I'm right where I need to be. Have any of you thought about New Mexico??? Just within the past year, it has occurred to me many times why this is such a good choice for retirement. We do not have hurricanes, we do not have tornados (okay, maybe rarely), we are not prone to earthquakes, the weather is jolly darn good 90% of the time (spring is out - way too windy) and we don't have more than a couple or four inches of snow in the winter. We don't start our furnace/heater until November and it's only in use until maybe early April. The air conditioner was just put to use two weeks ago and we won't need it past the first part of September. Low cost of living, lots of homes (reasonably priced - info upon request) for sale and the number of things one can do in New Mexico are practically endless. We have everything but a beach (and if California keeps shaking, we might have beachfront property - not that I'd wish that on California...). We have skiing, a big lake (fondly called Elephant Butt [Butte]) for water activities, stream, river and lake fishing out the kazoo, mountains to climb, white sands in which to wallow, beautiful sunsets and terrible drivers. Oops - that one slipped out! The margarita (and Bud Light) are the State Drinks (if one is so inclined) and this is the Land of Manana (read it like Spanish). Manana is way much better than I needed that right this very minute and no excuses! Shaded patios, cool evening breezes and gorgeous cool summer mornings (at least until 7:00 a.m.). Besides, I need a replacement beginning March 1, 2012. Brand new lab, tech-designed, bright and LEED, tons of space, a separate storage room for blocks and slides AND a volatile storage room with two acid cabinets and two xylene/alcohol cabinets and a salary (that's another subject, I do work for a State...). Can't have everything, but this is darned close. And I do not work for the Chamber of Commerce! [Histonet] 10% NBF Shelf Life?
I have just come from the Histonet Archives (cough, cough...) and my Extensive Library of Histology Reference Books (Sheehan, Carson and Lillie) and have found nothing on the supposed or generally-accepted shelf-life of 10% NBF. Is there any such number? For example, we make our own 10% NBF in 20L carboys; our usage varies from day to day so sometimes it's used quickly and sometimes it sits for several weeks. It has been proposed to me that we add shelf life date to our carboys when a new batch is made. I know there is someone out there that has investigated this before and can give me a time and a reference (and I need the reference as well for completeness). Thank you so much - I'm standing by. Funny you should ask... just last week I had two requests for a Von Kossa. That's when I found out that, despite the fact that we just moved into a brand new building with all the bells and whistles, there was not one single UV light in any of the many hoods that had been installed. Heaven forbid I could even find an incandescent bulb (we are a green building). So, I prepped my slides, put them in a clear glass Coplin jar and parked the jar on the hood of my car for an hour. Works like a charm. This all depends, naturally, on (1) if you even HAVE sunshine where you live; (2) how far a walk it is to the sunshine, and (3) whether you have a car hood on which to park the Coplin jar. Needless to say, out of the line of sight of curious onlookers with sticky fingers and no business wondering what that glass jar is... Nonetheless, it works just fine! Deparaffinize and hydrate to DH20. Place slides into 0.25% potassium permanganate for 30 min to 1 hour (visually inspect to measure level of melanin removed); wash in running tap H20 for 1 minute and place into 5% oxalic acid for 2-5 minutes until section becomes clear. I agree - I have a totally Leica lab except for my Sakura tape coverslipper and I would not trade it for anything. I test-drove the Leica glass but it seemed that there were more minute (my-noot) adjustments than for the Space Station and I could see problems down the line (sorry, Leica!). I know some love tape and some hate it but it's the absolute easiest to use and adjusting for drip (xylene) and length of coverslip (for those pesky Ventana-labeled slides) is a breeze. My bosses have no issues with optical quality or photography with the tape, either. This is New Mexico where humidity is a rumor. The humidity in the lab here - as I write - is 18% and that's on a really wet day! If I have static issues with my ribbons, I just lean a little bit toward the block and breathe on it and the ribbons just float (in a good way) off the knife. I do that so often that when I use my sewing machine, I find myself breathing on the material. That's just sad! But try the Breathing Thing. Or not. For those of you who work in a veterinary diagnostic lab where there is a BSL-2 and/or BSL-3 necropsy room, I could use some assistance with designing some SOPs for maintenance of those rooms. For example, what are your daily routines and/or procedures to ensure that those rooms are clean, temperatures checked, setup requirements met, and so forth. This is not writing pathologists' SOPs for specific necropsy procedures, but more the day-to-day operation and maintenance of the rooms in which a pathologist would work. Gremlins. That's what it is. I recently cut in a case with two parts - one Extremely Thin Black Crescent of tissue in one cassette and the other tissue in the next cassette. The next morning, the Extremely Thin Black Crescent was NOT in the cassette. Gulp. Sometime later, as I was almost done embedding, I opened a cassette and there - lo and behold! - was the Extremely Thin Black Crescent. Creepy. I had the pathologist confirm that it was, indeed, the Extremely Thin Black Crescent of tissue that had started out somewhere else... [Histonet] I'm in the New Lab!
We are physically moved and Organized Chaos prevails (except in Histo, of course!). There's been some discussion about what would be needed when designing a new lab and I agree with what has been offered - and actually used this sort of input from Histonet, along with the lab design course at NSH (which was priceless), and visiting a couple of labs before meeting with the architects. It was a long and arduous process but, with the exception of the chairs chosen by the majority in this building (about 700 people), was absolutely worth all the pain and agony. Please make note of our new physical address and a direct line to the histo lab (which I'll answer just as soon as I figure out how to work all those buttons and knobs!). We're getting ready to move into our new building (YAHOO!) in early September - finally. In planning where we will store our cases for the required 6-week retention time, I have proposed that the tissues (in 10% NBF) be shelved in the walk-in cooler (4 degrees C). Is there any reason tissues-in-formalin could NOT be stored refrigerated for that length of time? It is extremely rare that we are required to pull and recut wet tissue, but that possibility always exists. Thanks, everyone. I went to eBay and found a set of three (total $12.00) stainless steel food prep trays, roughly 9x5x2 (LxWxH). I fill them with DI water and freeze them. I also inherited a larger stainless steel container that's 12.5x10.5x4.5 - same deal, DI water and freeze. I face blocks, put them on the ice and add a little DI water and soak for 10-15 minutes - blocks cut like a dream! They stay frozen for a l - o - n - g time! Try a local restaurant supply. [Histonet] Waterbath Options
Okay, time for my two cents' worth. Water baths - I don't like the glass-dish-insert kind because water can get beneath the glass dish and can temp fluctuations. And if the glass insert breaks, it might be difficult to find a replacement (Pyrex?). I like the good ol' fashioned round or rectangular black-lined fill-it-with-water-and-turn-it-on kind. They hold temp much better and because they are lined with something like black Teflon (or whatever), they clean like a dream. I found that I really do not like the romantic under-tissue lighting. It's all about preferences and this is mine...not that you asked, but that never stopped me before, did it? Happy Tuesday! [Histonet] Storage Boxes
One of my former labs used plastic stacking boxes for holding blocks between cutting and filing, as tool trays and for many other purposes. These are the same boxes used in eyeglass fabricators (LensCrafters, etc.). I've been searching for them for quite some time and have just found a supplier at www.westernoptical.com (and likely other optical suppliers as well) - search for plastic stackable shop trays. I just bought a dozen for $30. I find them indispensible, which is why I've been looking for them for so long. I have my processor set to notify me at 1500 blocks, at which time I change the paraffins. I usually run 500 in the other solutions (same notification system) before changing. It works for me! Salutations (that's my real name) to all of you in the VETERINARY DIAGNOSTIC world. I need to come up with the cost of producing one HE slide and because I'm Old and Lazy (not), I'm wondering if any of you have already done this complicated calculation and have a number I could use. I barely have enough time to gross in, type reports, embed, cut, stain, coverslip, file slides/blocks and do special stains and immunos as it is! I shouldn't claim I'm lazy but why invent the wheel twice, I'm sayin'. I'd appreciate any cost you might have... Muchas gracias! [Histonet] Accessioning Similar Samples
Regarding the need to avoid accessioning similar samples consecutively, I addressed this last week with my bosses. Specifically, I asked that spleen specimens not be accessioned consecutively (because of shattering and static and the possibility of fragments being carried over); my response was that I should just not CUT the blocks consecutively. Not the point I was trying to make but typical in some cases when addressing the issues histotechs face, true? If it weren't for a med-tech-turned-pathologist that took me up on my interest in becoming trained in histology (1967), I would not have just marked my 41st year as a histologist. Back then, you just had to have OJT and study independently for your written and practical tests. I had no idea what I was getting into but it was one of the absolute best choices I ever made.
