[Histonet] PTEN Question (Evan Solanki)

I am by no expert in PTEN expression but we do a fair amount of antibody
workups.  For new antibodies that we don't have known protocols and or
types of tissues we refer to "the human protein atlas".  This gives volumes
of information like both normal and abnormal tissue expression, and many
other facts that may be relevant.

We have this problem every year during the summer months.  Our fix is to
bottle all chemicals when not in use.  We have a plethora of 1 liter
nalgene glass bottles we use for chemical solutions.  It works for us.

We are looking at the possible use of microarrays or tissue rolls for
immuno controls on a routine basis. I would like to talk to people who are
currently doing their immunos this way.
We run about 120 slides a day with about 170 different primaries. Anyone
with previous experience in this area (good or bad) who has any
suggestions, I would be glad to hear about them.

http://jmdhistology.com/) that makes them.  They have fast turnaround times
of 2 weeks at $5.00 per core.  They use a new proprietary system that

Hello Tim,

I also have had this cracking problem in the past but since resolved.
I'm not sure about the specifics of the cracking problem but the cold
plate might have something to do with it.  Our cold plate is set to
-5C and use Paraplast from McCormick Scientific.  Our paraffin is
roughly the same melting temp as yours, so probably not much
difference.

Unfortunately trial and error is apart of our jobs since there is
relatively little uniformity in histology equipment and products. Have
you tried setting the cold plate to a warmer temperature yet?  It's
worth a try and if that doesn't work maybe the paraffin has been
getting too hot.  I know when the paraffin gets too hot, the wax and
plastic separate or break down and cause inconsistencies in the
paraffin.  To test this, take an external thermometer and place inside
the paraffin tank.  Then record the temp every hour for some time.
This will tell you if the tank itself is a consistent temperature.

Also, are the blocks taken off the cold plate and immediately jarred
loose from the mold?  Sometimes when I do this the shock of pried out
of the mold can cause the paraffin to become brittle and break rather
than bend.

ever had to cut, or were asked to cut?

For me, I work in a research facility where I do everything.  One day
a botanist emailed me to cut pine on the cryostat and asked what the
largest size should be.  I told him 2 mm x 2mm and explained the
process as he had no idea what I was going to do. I thought ok, what
is pine?  He brought 5 pieces of wood pieces (pine) and asked if they
could be cut cross section at 10um with 5 sections per slide.  Of
course this is not something I have ever had expience in doing, and
they are hard as a rock.  The trick that ended up working, was to soak
the wood in warm water for 30 minutes, embed in OCT and cut.  After
cutting the sections were all balled up, so I floated them on water
and picked them up on the slide.  The sections were still folded and
looked horiffic in my mind but he loved them.  He said was going to
look at them under a Raman confocal and wanted to see the plant cell
wall.

Hello Patrick,

We have this same cryostat except it's the CM 3050S.  Ours also has
this issue usually after it's been running for many months with a
defrost.  The typical cause of your problem is the cylinder on which
the block holder is attached has ice crystals or is frozen.  I feel
the cause of this is because the glass was left open too long while
cutting.  This allowed moister from the room to condense on the
cylinder and subsequently causes a stop go motion.

 Since I am in charge of all histology equipment maintenance, my
solution is to turn off the cryostat, take the microtome out of the
cavity and let it dry overnight.  Then in the morning, I physically
dry the microtome.  Using cryostat oil, oil all the moving parts
including the cylinder, then wipe off excess.  Before putting the
microtome back into the cavity, the cavity itself must also be
thoroughly dried.  The final cool down process usually takes about 1-2
hours.

Hello All,

Does anyone have a protocol for the Anti-Neutrophil antibody [NIMP-R14]
(ab2557) for mouse tissue?  I have been trying to extinguish the background
staining but cannot find the right combination.
​ We are doing this by hand. ​
 The recommended dilution for this antibody is 1:50 with HIER.   See used
protocol below.

So far I have tried:
1. concentrations 1:25, 1:50, 1:75, 1:100  - 1:50 gives very high
background but good positive staining, 1:100 gives low background but not
much positive staining.

2. HIER - 3, 5, 10,  20, minutes  @ 96C, 20 minutes at 60C  all give the
same results - too much back ground

3. We have tried using the H2O2 before the primary and after, increasing
the time from 30 minutes to 45 minutes.

4. Tried incubating in the primary for shorter time (20 minutes) and longer
overnight at 4C.

5. Tried blocking using 3 different serums, first each one then
combinations and longer times.

6. Tried cutting the DAB concentration in 1/2 then 1/3.


We have not tried enzyme or acid epitope retrieval yet.



*Does anyone have a working protocol or suggestions on what to try?*

Procedure:

Deparaffinize

1.  Heat slides to 60C for 10 minutes (oven).

2.  Dry slides at room temp 5 minutes.

3.  Xylene 5 minutes.

4.  Xylene 5 minutes.

5.  100% ethanol 5 minutes (dehydration).

6.  100% ethanol 2 minutes (dehydration).

7.  95% ethanol for 2 minutes (rehydration).

8.  70% ethanol for 2 minutes (rehydration).

9.   Run in DI water for 10 minutes.



Antigen Retrieval

1.  Submerge in 0.01M Citrate Buffer (pH 6.0) at 96C for 5 minutes.

2.  Cold running water 5 minutes.



Staining

1.  Wash in PBS for 1 min.

2.  Permeabilize with 0.1% Triton X / 2.5% horse serum + 2.5% fetal
bovine serum for 2 hours.

3.  Incubate in primary antibody 30-45 minutes at in 2.5% horse serum
in PBS.

4.  Wash 3x in PBS/tween for 5 minutes each.

5.  Fresh 3.0% hydrogen peroxide (H2O2) 2x for 20 minutes each.

6.  Wash 3x PBS 2 minutes each.

7.  Incubate with anti-Rat Ig impress solution (according to vector’s
instructions) 45 minutes.

8.  Wash 3x in PBS for 5 minutes each.

9.  Prepare Impact DAB substrate (see insert).

10.  Using a light microscope, incubate sections with 50-100ul, until
desired darkness occurs (5-30 seconds).

11.  Stop DAB reaction using deionized water.

12.  Wash in DH2O for 5 minutes

13.  Counterstain in Harris hematoxylin for 30 seconds.

14.  Wash in DH2O for 5 minutes.

15.  Dehydrate 2x in 95% alcohol for 30 seconds each.

16.  Dehydrate 3x in 100% alcohol for 2 minutes each.

17.  Clear 2x in xylene 5 minutes each.

18.  Mount coverslips using Leica mounting media.

I have done myeloperoxidase staining on animal tissue.  Is this the
same for plants?  Most of the plant antibody sites I have gone to do
not list peroxides as a test.

[Histonet] Neutrophil staining

sectioning of GMA blocks containing biomaterial devices and subsequent
HE staining.  The devices are cut for embedding to about 8mm in
length, and contain adhesives, thermoplastic membranes, various

samples to you, and would be looking for about 25-50 samples/month.

That depends on your protocol. Which protocol are you using and what
does your protocol say to use the schiffs at?  Consider that schiffs
is usually stored at 4C in brown or opaque bottles.  This suggests it
is light sensitive.  Our protocol say to use the schiffs at 37C for 15
minutes but I have used it at 22C for 30 minutes and at 60C for 10
minutes with consistent results.  We have found that using it at 60C
reduces it's shelf life considerably and do not suggest it unless you
are familiar with doing so.

Several years ago while in the beginning stages of setting up my
histology lab, I looked into this possibility (cutting at home).  The
town hall laughed and said it is illegal for a business to run out of
ones house that uses chemicals and paraffin because of it's flammable
and explosive properties.  This made sense, but I also had heard
stories of beginning companies who have done this.  Therefore a
business running out of a residential house is doing so illegally and
maybe without insurance.

Hello Deanna,

This sounds like BS to me, maybe just a idiosyncrasy with their
management.  They are probably set in their ways and refuse to change
or believe in another way to do it.  It might be that one person once
had artifact and it caused an uproar for the pathologists who in turn
implemented this rule.  I know, that if some kinds of tissue is soaked
too long, the tissue can bubble up out of the block and be rendered
unusable.  If I were you, I would just go along with their way of
cutting and don't try to change them.  Change is only possible if
someone is open to it.

I agree water is always a potential issue.  Tissue can be finicky though.
 Have you tried poly-L-lysine coated slides?  They do help is some
occasions.  If you need a protocol, let me know, I can write one up, very

[Histonet] Does Formalin fixed frozen sections for antibodies need to be kept frozen?

Hello I am working at a research facility that has thousands of formalin
fixed frozen heart sections in their freezers.  They have been led to
believe that this is necessary for their antibodies to work.  I do not
believe this is the case.  Does anyone know of antibodies that will lose
their antigenicity if stored at room temp?

bone samples, data needed is % of new bone present in samples from sites
grafted with freeze-dried mineralized cancellous allograft bone.  We can do
the plastics on it but the computer assisted part, I do not know anything
