I just took (and passed!) the QLS exam this month. My test included several
questions about needle stick injuries, chemical spills and safety, and fire
safety.  The list they have is a great start for studying but I would also
get the books they recommend. I admit the test was more difficult than I

The kind with the blade was damaging our floors and the blade would break
causing a major safety issue. We just bought a long handled scraper from
Bel Art; it works really well.

We are also going through humidity issues post inspection. Our values are
based on the specifications in the equipment manuals and are 30%-80%. We
use a digital reader. We have been dipping below that and are working to
get the humidity up. We have brought in a small humidifier but it is not
keeping up on very dry days.
It is in our policy to report out of range values to our supervisor and she
is now looking in to a more permanent solution.
We would also appreciate any comments on how other labs have increased
their humidity.

We also received this notice. We contacted our local CLIA office who had
heard of it this week as well. We are a small lab, so we are not sure yet

love it. I had so many problems with the Leica (which can be attached to

I also had great results with American Mastertech's stain kits and agree
that their customer service is great. I've ordered HP control slides from

CLIA requirements are very clear for any testing personnel doing the
grossing. Be sure you read the CLIA law regarding grossing requirements and
be sure all of your grossing personnel not only meet these requirements but
that their qualifications are well documented-including updated resumes
indicating that they are CLIA eligible to gross. All grossing is a high
complexity task.
I would highly recommend you review the CLIA website for yourself and if you
have any questions, call your local CLIA office. I just went through my CLIA
inspection two weeks ago, feel free to contact me if you have any other

[Histonet] Quick Drying Mounting Media

Hi histonet,
We have a brand new Leica CV5030 Coverslipper and I am looking for the
quickest drying media available. We are currently using Micromount by
Surgipath/Leica and it is not drying fast enough.
Any suggestions are appreciated as we have to package our slides for
shipping and the media is either sticking to the bubble wrap or creating

Hi Nicole,
You must get a microscope. Spend $600, get a teaching binocular one. You
absolutely can not have a histology lab without a microscope. You must be
able to check the quality of your staining and cutting.
That being said, the lab that does the PC component should also do QC on
your slides. If there are major issues they should address them right away,
otherwise, you can set up a QC Form that they fill out quarterly to assure
your slides are meeting their standards.

 We run an appropriate positive control on every patient test slide to test
 that the antibody and all components used on that slide worked.  This may
 be overkill but it helps us ensure that each slide worked properly not
 just the run.

We do all of our grossing and use the CLIA regulations:
TESTING PERSONNEL (42 CFR 493 1489)
1. Licensed MD, DO or DPM.
2. Doctorate, master's, or bachelor's in laboratory science.
3. Education and training equivalent to an associate degree in a
laboratory science or medical laboratory technology that includes at least
60 semester hours including 24 semester hrs of medical lab technology and
at least 3 months training in each specialty in which high complexity
testing is performed; or, 60 semester hrs including 24 hrs of
science that includes 6 hrs chemistry, 6 hrs biology, and 12 hrs
chemistry, biology or, medical lab tech in any combination and laboratory
training that includes either: completion of a clinical lab
training program and at least 3 months training in each specialty in which
high complexity testing is performed.

It is my understanding that if there is no cutting involved (i.e. counting
and measuring GI biopsies) it is not considered a high complexity task
and anyone in the lab would be able to gross in that regard.

[Histonet] parasites in stool

Hi all,
I have received some parasitic stool samples, one fixed in formalin, the
other in PVU. My tasks are:
1) to try to remove as much debris from the stool sample as possible while
keeping the parasites intact (I need to try to filter it without harming
the very fragile organisms)
2) once I am able to separate the organisms, I need to stain them. We have
tried using a Gram stain and it does not work, we are thinking we should
try a Giemsa and an HE.
If anyone has ever worked with anything similar, I would really appreciate
some feedback, suggestions, advice with how to filter these samples and
stain them.

We use the AmericanMasterTech Alcian Blue pH 2.5 and use this procedure:

Place slides in 3% Acetic Acid for 3 minutes.
Place slides in Alcian Blue for 30 minutes.
Rinse slides in water
Place slides in 0.5% Periodic Acid for 10 minutes.
Rinse slides in water
Place slides in Schiff Reagent for 15 minutes
Rinse slides in HOT water for 5 minutes.
Counter stain in Hematoxylin for 2 minutes.
Rinse slides in water, Dehydrate slides through Absolute Alcohol, Clear
through Xylene

Yes, you are correct. One year OJT plus a BA/BS with required # of
biology/chemistry credits for HTL.  When I tried to call to see if I
qualified they said that they can not tell you over the phone. You have to
apply (and pay the $200) to see if you qualify.

 You might be eligible to sit for the registry already either HT or HTL,
 you just need to have the correct number of credits in biology and
 chemistry to meet the requirements.  I would call ASCP to see if you
 have the requirements, it seems to me that you have the experience
 needed you just need to make sure you have the credits.  That's my
 opinion.  All you need is one year on the job training with a BS or BA
 to sit for the HTL.  I think I'm correct on this, but if I'm not we'll
 know soon enough from the replys.

[Histonet] Marginated Staining on IHC

Hi histonet,

I am back with a problem I've been having with my IHC slides.  We had a PM
recently and had thought that would solve this problem, but as I do more and
more slides, the problem remains.  I am seeing marginated staining at the
edge of the tissue on the controls as well as the patient tissue.  This
occurs at the top, middle, and bottom of the slides.  This occurs with any
antibody. This does not occur with every run, but the more slides I have on
the stainer, the more frequently it occurs.  I have contacted Thermo, who
now takes care of our old R.A.S. Microm HMS 710i, but I have further faith
that the histonet can help me.  I've seen similar posts in the archives, but
did not see any responses.

[Histonet] IHC Stainer: Microm HMS 710i

I am looking for help/advice for using our automated immunostainer the
Microm HMS 710i  .(Our user manual says Richard Allan Sci and the labels on
the side say LabVision, we have calls into Thermo's LabVision department but
they are unsure if they can assist us)

Specifically, one of our problems is that some of our stains are producing
dark coloring around the edges of the tissue. I am having issues
troubleshooting and absolutely can not change any of the programming.

I have been using this stainer independently for over a year, taught to use
it by someone else who was taught by someone else. I am looking to change
some programming as well as troubleshoot. Anyone who has experience with
this stainer is welcome to contact me.

I am also about to take my HTL exam. The website has all the information you
are looking for.  I've been through these testing facilities when I took my
GRE. Make sure you have at least 2 forms of I.D. with you, do not bring a
cell phone, etc (all this is on the ASCP website).  You will not get any
results right away.  I don't remember if you can go back and answer missed
questions, but I doubt it. The way it works is that there is a pool of about
500+ questions worth at most 900 points.  Each question is worth a certain
amount of points based on the difficulty of the question. If you get a
question correct, the next one will be harder. If you get it wrong, the next
will be easier.  You will answer 100 questions.
Surf through the website and you will find all the answers you seek.

Well, I'm getting close to my HT exam date and was wondering if any of

itself as far as if it is weighted or just a straight 150 questions. Can

you have passed or not?  I've been told in the past that the computer
would just shut off and a screen would come up pass or fail.  Anyway, I
have been focusing on Frieda Carson as well as the AFIP manual.  Any
suggestions for a successful passing score?  I think you have to get a
minimum of 400.  Should I be concerned with microwave special stains?  I
would think the test is more old school.  Any helpful hints would be

[Histonet] SAFE-CLEAR Xylene Substitute

Our pathologist has agreed to let us try out SAFE-CLEAR xylene substitute in
our VIP processor.  We currently have 2 changes of xylene at 30 minutes each
and process mostly small surgical specimens.  

Do I need to increase the time the tissue spends in the SAFE-CLEAR?

I have searched the Histonet Archives for answers about xylene substitutes,
and have mostly found what I am looking for, but am unclear on whether or
not I need to increase the time during processing.
