We have the VCP-3001 cassette printer and we buy the printer ribbon from Leica 
(800-225-3035).  The stock number is V0590001F and it cost us $83.35 per ribbon 
which we bought several years ago.  I don't know if it is the same one for your 
VCP-2001 but I'm sure when you call them, they can help you.

 Some people have issues with the smell and the other option is Anatech ProPar 
 or Richard Allan Thermo non-aliphatic hydrocarbon subs which have very low 
 odor and are also about the same price.  We can't use the orange smell in our 
 lab as some people have allergies and others just don't like it.  Both work 
 types work very well and it is a lab choice for the options, not a quality 
 choice.  Be aware most processor companies still require xylene in the clean 
 cycle to keep your system healthy.

 Histo-Clear can be directly substituted for Xylene at every point in 
 processing and staining.  It's completely non-toxic (even has a food grade 
 rating), is made from oranges, and smells delicious. You don't have to work 
 in a hood and it seems to make tissue less brittle than Xylene also. Some 
 guys in our Histology Core told us about it and we have been happy with it.

We have the B/R 9700 ProCycler to recycle alcohol, xylene, and formalin.  The 
Advantage must be a newer model.  About 70 gal of alcohol, 40 gal of xylene, 
and 20 gal of formalin is recycled every 3 months.  This recycler is easy to 
use and works well for us.  As with any equipment, you need to maintain it by 
doing pm checks (replacing parts) as needed.

 experience. I have known some Pathologist to read the mucin that way

 Hello all,
 
 I have a request to carry out a combined staining with HE + Alcian Blue 
 pH2.5.I have tried in vain to get this to work but regardless of the 
 order of staining the end result is dark blue/purple mucin.  I have 
 carried out a parallel experiment whereby the staining has been checked 
 microscopically then stopped after each of the dyes.   (Thus giving 3 
 slides stained with:  AB, AB+H and AB+H+E)  The AB and AB+H are really 
 nicely stained but as soon as the eosin is added (using 2 different stains 
 and a variety of times) the mucin staining looks similar to the nuclear 
 stain.  I am expecting the alcian blue to remain turquoise: should it?
 
 For information, I am trying to stain salmon intestine at 5 microns for 
 using the following:
 
 Alcian Blue 8GX (certified), pH has been checked
 Haematoxylin Z (Cellpath Uk)
 1% aq Eosin (Cellpath uk) and lab prepared solution from dye.

We just obtained a VIP 6 processor and we have a program for routine overnight 
processing.  It seems to work well.  I will send or fax it to you by Monday.  
The Tissue-tek sales rep that installed and trained us, mentioned about 
increasing the times in the alcohols for fatty/breast tissue.  You can try this 
and make any adjustments to the program I am sending.  We have two stations of 
formalin at two hours each.  As a side note, we have two Excelsior processors 
also and except for a few glitches, they seem to work fine.  Take care.

Tasha,
We have the Excelsior which we received a few weeks ago.  We have not used it 
yet for patient specimens but will very soon.  Our test runs went well except 
some breast tissue (probably cut too thick).  Overall, the pathologist was 
happy with the results.  There are many good features, but one of them was the 
potential savings of alcohol solutions.  It has a built in hydrometer that 
measures the percentage of alcohol in the containers and it will let you know 
when to change it.  We are also looking into the Tissue Tek VIP.  We have the 
300 model which we like but it is nearing it's life expectancy.

 Sigma E-2886 extravidin peroxidase, 1:1000 in 0.1 M PBS or sodium 
 acetate on almost every IHC variation (fixation, species, primary) we 
 do.  Supposed to be less nonspecific binding than streptavidin.

 Greetings to all histonetters- I have a pathologist that wants to show fibrin 
 in lung. She wants also black nuclei and we tried  a Movat's to look at 
 fibrin. The fibrin in the lung is showing up light blue not red!
 Any suggestions on how to show fibrin distinctly, with black nuclei?

 We no longer release specimens to patients.  If they want their tissue they 
 have to go through a funeral home.  Here in CT, funeral homes are the only 
 agencies licenced to bury or incinerate human tissue.

 Hi Peggy,  From my experience, during the decal process the cement will fall 
 out or loosen enough so that you can gently remove it and maintain fibrous 
 ingrowth and detail. I have removed ceramic; implants; stents and cement 
 using a scalpel and then embedded in paraffin for cuts on a sledge microtome. 
 The detail is beautiful even without the cement present. If you have the 
 capability, a corresponding section in PMMA can be processed.  ~ Vicki

 Does it have to be a coverslip?  There are some mounting media that harden 
 and form a barrier with optical qualities similar to that of glass.
 
 It might not be a perfect solution, but it might work.
 
 I think the stuff I used was called Crystal Mount (?).

 Yes, the scintillation vials are notorious for shattering in the freezer.  
 However, if you put them in a plastic container with a lid on, the shattered 
 glass will be contained and not all over your freezer.  When breaking larger 
 glass containers, again place them in the freezer, remove from the freezer 
 after 20 minutes or so, take the lid off, wrap jar in paper towels and hold 
 the ends closed.  Wear glovesand eye protection in case shards of glass 
 escape.  Strike the jar with a hammer and unroll the paper towel over a 
 garbage can.  Rinse the block in tap water to remove any small shards of 
 glass.  This is a great way to vent frustration!!
