Hello Histonetters,

What is the take home message of this revision? How are other labs complying 
with respect to Special staining?  Do you keep records of individual case 
control verification or just individual block verification? Does anyone run NEG 
controls for Special staining? Is this regulation requiring Neg Controls for 
special stains? Any thoughts are appreciated.

[Histonet] Syringe Pump error Ventana??

[Histonet] histology AND incinerators?

forceps? We are setting up to begin using Germinator 500 at both gross and 
embed center, was wondering if any other labs have used this type of

[Histonet] New Cap cross contamination at gross bench

Anyone out there see the new CAP requirement regarding cross contamination at 
gross bench and cleaning of forceps.

How are other people planning to comply. My supervisor has instructed me to use 
a Germinatior 500 at gross bench and at Embedd center.

I have no experience using these sterilizers but my initial thought is that I 
will need to be really careful not to inadvertently apply too much heat to the 
tissue.

Any thoughts appreciated,

I wish you had contacted me a few months back. Management moved a tech over 
internally here.

I will put you in contact with White River Junction VAMC . They may have a 
postion open. Also, I would check University of Vermont Med center;, 
Brattleboro Memorial hospital, Dartmouth Medical Center, Catholic Medical 
Center, Manchester.

I know people at all these locations, often times a latent position (everyone 
knows about it ) won't post until a certain period so you wouldn' t know its 
there without asking. That said, Ill ask around.

Hello Kelli,

You may remember me as one of your online graduates from the GC histological 
science program. How are you? For some reason I thought your involvement with 
the GC histo program had changed, but seeing your recent post to the histonet 
it appears your still the director of that program?

I hope things are well. You were instrumental in helping me complete that 
program successfully and were also a good instructor. Ive been at the VA 
Manchester NH for 2 years now. I am the lone histotech here since the other 
technician left. Things are going great though we have a critical need for  
another qualified technician.  Please let me know if any of your students 
especially those already holding  a BS would be interested in employment at the

[Histonet] recycled alcohol versus reagent grade alcohol

Hello Histonet,

How do other labs test and verify the percent grade of alcohols and what is the 
assumed percent grade of reagent alcohol? We use recycled alcohols but today 
based on not having reagent grade we used 97% tested recycled as our flush.

The reasoning behind this was that Reagent grade varies from 95%-greater than 
100%. Is this true?

[Histonet] sections blowing apart in stainer

Hello,

Cut some skins the today (3 sections one slide) and the top two sections seemed 
to blow apart and drift  in the stainer (not on the bath). The bottom section 
looks good.

This is strange as  I have seen the bottom section move as a result of water or 
not adhering to slide. However is in this case the top two sections appear 
blown apart and drifted and this occurred while staining?

Any thoughts appreciated.

That's interesting, I didn't realize gelatin is the water soluable protein of 
collagen. No experience with staining gelatin, but have you considered 
MassonTrichrome. 

It is used to differentiate collagen from smooth muscle...

You need to explain treated tissue. 

Gelatin is collagen that has been boiled until the protein has lost all its 
fibrous nature and changed into a water-soluble protein. Gelatin is made 
permanently insoluble by adequate formaldehyde fixation. It is stained by 
anionic dyes (such as eosin in the HE method), but it does not show as fibres 
when you look at the section or smear through a microscope. 

If this doesn't answer your question, please explain your problem and involve 
your boss in future email exchanges.

I use a block trimmer. I think it is safer for wrists. Being that you push the 
block against it and I seemed to always pull when I scraped blocks.

I am guitar player and need my wrists. 

I don't think it is a strong argument to say its faster then hand scraping 
atleast not until a good technique is developed by the tech.

 after embedding,  we purchased a paraffin block trimmer.  It saves 
 time and the stress on finger joints compared to scraping cassettes 
 daily.  No matter how careful we were during embedding to keep excess 
 paraffin off cassette edges,
 we were never successful.   Several vendors have these and you may be able
 to find a refurbished one.

for the whole bee I probably would process and embed it in glycol methacrylate 
(gma) it is much harder and would give better sections, we have done zebra fish 
and several other harder tissues including calcified bone in GMA.

 wanted to show the difference between the stingers.  I wasn't sure what to do 
 so I processed and handled like everything else.  I was able to get some good

 there should be at least one good stinger in each block and it worked.

Speaking for myself and the labs I have managed, I have been checking ALL temps 
and recording them since the early 90’s. These include waters baths for tissue 
flotation, and ones used for special stains, ovens, refrigerators, freezers,

I agree with Dawn . Basically use a numeric for the Block and an Alpha for the 
specimen. This way a large resection specimen would receive an A and all blocks 
for said specimen would be labeled as A1,A2,A3...ect

We use a numeric for the accession number (S-14-1234), followed by alpha for 
the specimen(A, B, C), and within that have numeric for the number of blocks in 
each specimen (A1, A2, A3). We haven't ever gone to Z in our current LIS, and 
the ability to add unlimited blocks to each specimen makes it easy to deal with.
