Hi Chris, CAP actually has a specific checklist question on Negative Controls. The question is ANP.22570 and if I remember correctly it is quite extensive. Most of the CAP questions are about 7 to 10 lines. The Negative control question is more than a page long. Get your hands a copy of the most current checklist for anatomic pathology - That will include Immunohistochemistry. By the way, the rule is that a negative control must be included with each run and there is much, much more. We do NOT run another negative control if we have to repeat one of a group of immunos or if a pathologist orders additional stains at a later point. I am wondering if this is good practice, and acceptable with CAP. I would love to find some literature that could help me make a case either way on this. Actually, any literature or pointers regarding IHCs and becoming CAP accredited will probably help save what little hair I have left. When I asked them that question, they said that it was usually at the discretion of the pathologist in charge. Hi Donna, I use Dako's Myo-D1 cat. M3512. Use it at 1:25 I use Dako's labeled polymer kit, Envision + Mouse. Use Trilogy (from Cell Marque) in a steamer. That means: Start the steamer and put a two Coplin jars in filled with Trilogy. Place all your slides in 1st Coplin jar for 30 min. right then, at RT. The temp will go up to 95 degrees as the Trilogy acts as a deparaffinizer. Then put he slides in the 2nd Coplin jar for 30 min. and Trilogy acts as AR. Good luck. ANTIBODY LOTS I have a Dako and I have a Bond too. For the Dako I use my positive control. I use 3 slides. One is treated as the negative control - no antibody One is treated with the current lot One is treated with the new lot. I keep all QC slides together. I keep the 3 slides together. For my Bond, I use 2 slides of my positive control When a new lot comes in I run the new lot and the negative control. Then I compare them to the previous lot that I ran in the past. I keep the slides in chronological order - the previous lot goes back with The slides from that date. The NEW Lot slides get filed with today's date. DETECTION SYSTEM LOTS For the Bond I QC the detection kit using an antibody like Ki-67, it works well and always shows good nuclear staining. I indicate the received date, and QC date. I run the QC on my Ki-67 positive control. I use 2 slides. One is treated as the negative control - no antibody. One is treated with Ki-67. I compare these to the previous Detection kit QC slides I created a written procedures for all of the above. Hope this helps. Yes, you should run negative controls with each run - today and tomorrow. Something can happen in tomorrow's run so tomorrow's negative control will have different issues from today's negative control. The negative control should go through everything that your slides go through, whether it's today's great run or tomorrows electrical black out or whatever mishap. Whenever you run a test you must have a positive control and a negative control. Hi Cindy, That too has happened to me and the process that you do to remedy it, works for me too. (take off coverslip properly, re-apply DAB, and you get a reaction.) My experience is that the slides are not shot, as described by another Histonetter. There was definitely something else going on. I work with the Dako too. I run IHC on the Dako autostainer. On one of our runs there was not enough DAB to dispense on all the slides. My tech dehydrated the slides and cover slipped them. We printed the stain log on the run and confirmed that the DAB was not dispensed on three slides. The cover slips were removed and the slide were soaked in xylene to remove the cover slipping media. The slides were rehydrated and DAB was applied. There was still no reaction after 10 minutes, The DAB was reapplied for up to 30 minutes with no reaction. Can anyone out there give me a reason that the slides would not react after the reapplication of the DAB. I have done this before when the DAB solution was short on the run and it worked. I can't figure out why this didn't react. I called tech service at Dako and talked to the supervisor, she agrees with me that the reapplication should have worked. I checked the run log and the only missing step is the DAB. The antibodies with the missing DAB have their own controls on the slides. Any thought out there as to what the problem is. I have had the PM done on the Dako which should resolve the problem according to Dako. There has not been a problem since. This mystery will drive me crazy until I can figure out why. Hi Michele, Yes, it's true, only MTM labs sells it and they Sell it as a kit and they also sell it along with a negative control reagent. That's how I buy it. They have a license or something and no else can sell it now. Their cat# is 9518 and I think it's called the CINteck Histology V-kit, (you should double check with them.) They will make you open up an account...
