Joanna, If it looks like it is under the section, then I would suspect  trapped 
reagent in air bubbles, folds, etc. If it is on top of the section then maybe 
more rinsing is needed.

Hi, I was hoping you guys can help us with our problem with modified gomori 
trichrome stain for frozen muscle biopsy. We've been seeing a lot of red 
blotches on our slides. We've tried 3 different manufactures of chromatrope 2 R 
dyes, including changing our recipe. So far no success. 

Hopefully  you can give us some suggestions.

Joanna, I agree with Tim... We also see this at times if the tissue has 
freezing artifact.  Gomori Trichrome is very unforgiving :)

Hi,

you can try, if you do not already, to wash briefly 
in a solution of 0.2 ml. of glacial acetic acid in
100 ml. of distilled water, before the final dehydration.
That makes the colors more delicate and transparent.
The Gomori's trichromic results are similar to those obtained 
with the Masson's trichrome staining:
Cytoplasms from bright red to greenish.

[Histonet] Gomori Trichrome and nemaline bodies?

Hello All,

 

I work in a core facility providing laboratory services to researchers.
I had a request to cut some frozen sections on muscle tissue, and stain
them with Gomori Trichrome, a technique we use routinely.  I did the
stain, checked them microscopically andconfirmed that the collagen
fibers were nicely stained.  But the researcher came back and said that
the slides did not show what she was looking for, which was nemaline
bodies, rod-shaped structures indicative of a disease called nemaline
myopathy.  I had never heard of this but went online and found several
articles on the disease, a couple of which showed images of nemaline
bodies stained with a modified Gomori Trichrome procedure.  But no
indications of what the modifications were.  Is anyone familiar with the
staining of these bodies, either using a modified Gomori trichrome, or
some other method?  Thanks.
