[Histonet] H Pylori IHC

Hello, we have just purchased the Leica Bond Max IHC stainer and love it. It's 
easy to use and produces beautiful stains.
However, Helico Pylori has been giving us problems with weak and incomplete 
staining (not all of the organisms staining).
Is there anyone using the Bond Max and successfully staining H. Pylori?
 We have tried two different antibody vendors, Leica and Biocare Medical giving 
similar results. We have also tried several different Antigen Retrieval methods.

[Histonet] H. pylori stain

Hello histoland

We currently use Ventana's h. pylori antibody on our Benchmark ultra.  Over the 
last few months our pathologists have been noticing "junk" that is staining 
with this antibody.  It seems to be on the tissue more than in the tissue and 
its not on all slides.  WE have tried changing our waterbath from tap water to 
distilled water with no difference in staining.  We thought maybe our paraffin 
was bad so we stained a "blank" block of paraffin but could not find the 
culprit...Any insight would be appreciated!

[Histonet] H. pylori by IHC

Recently someone placed a query regarding the precipitate with Cell Marques H. 
pylori marker.  The response was to filter it before use to clean it up.  May I 
ask how it is being filtered?  We have the same problem and it makes the slide 
difficult to interpret.

Hi,
 If you are looking at the possibility of doing IHC manually, I would
really recommend the Shandon Sequenza. It is a box that holds clips that
attach to your slides. The slides are held vertically and you drop reagents
into the top. The reagents replace the previous reagent which drips into a
catch tray below. It makes it **really** easy to reproducibly do IHC
manually.

We do HP and Mart-1 testing in house by hand using concentrates.  It's a pretty 
basic set up using a pressure cooker, stain tray, pap pen, and the reagents. At 
this point in time it's less expensive than automation.  We start them around 
10 am and finish around 12 or 1 pm.

Giemsa regressively for those. Not as sensitive as IHC. For IHC we used
Cell Marque by hand (with great results) and now we are automated.

If you do it by IHC it is the normal 2 hour or so process step-by-step.
Giemsa is quick 15 minutes or around there? Again, not as sensitive but
works.

We use Ventana's HP antibody, and we do see some of the small, non-specific 
globs of stain, but the pathologists are comfortable differentiating between 
the true and false staining since they are familiar with this artefact.

I haven't used HP, but have noticed this stippled effect especially when using 
40X on the microscope and looking at slides labeled with an Ventana alk-phos 
kit. I seem to have solved the problem by making sure no trace of H2O is left 
on the slides. It helps after washing slides to run them thru a few changes of 
alcohol then one acetone and then a couple xylene before coverslipping. Ventana 
suggested the acetone step. Tom T

have tried many, but our pathologists complain that they are not specific for 
H.Pylori and that all gram negative bacteria are staining.  Currently we are 
not running this but our pathologists are interested in it.

We use the Rabbit Polyclonal H. pylori from Cell Marque on the Ventana
Ultra's... We're a natinowide reference laboratory and our clients are all
very happy with the specificty of the staining.  Let me know if there's any

[Histonet] H pylori control on ventana immuno stainer

I am setting up a new lab and I am using the Ventana immuno stainer, does 
anyone know of any good HP controls I can purchase for now that will work well 
with the Ventana?

[Histonet] H. pylori RTU antibody

I was just wondering if a lot of people use the RTU antibodies available out
there for H. pylori.  Currently, we're testing the RTU from Dako and we're
having issues with a lot of background staining.  After tweaking the
protocol a lot, I've managed to get a lot of it removed, but there are still
some issues.  I was wondering if most people are making their own dilutions
to get a better stain or if the RTUs are commonplace.  I appreciate the
feedback!

Use the RTU from Biocare.  It is clean and easy.  You can use it with the Dako 
Flex Kit.

Hi,
 I am sure this will work. Granted I haven't tried it... The antibody is
directed against the bug, the H. pylori bacteria, not against a product of
the human it exists in. So it shouldn't matter where the bug is, it should
have the epitope the antibody binds with. I do Hepatitis B on non human
tissue all the time and it works fine.
