[Histonet] Leica Bond Max - non OEM products?

2019-09-05 Thread Dr. Stephen Chang [UML] via Histonet
Hello,

I was referred to join this listserv due to a question we have about our
Leica Bond Max. I work in a medical diagnostic laboratory. Does anyone know
where we can get good quality but cheaper reagents to use on the Leica Bond
Max, perhaps from a third party? The prices from Leica directly are somewhat
pricey and the person who referred me said she believes that many labs use
"aftermarket" supplies. I'm talking about epitope retrieval and wash
solution, etc, not antibodies.

Thanks in advance!

Sincerely,

Stephen W. Chang, MD, MBA

Hello Histology World!!!

Can muscle placed in Gluteraldehyde be processed for structure? In this
situation, all tissue was placed in Glut and we need to perform Enzyme
Histochemistry but we have no fresh tissue.

Thanks for your help.

Re: [Histonet] Leica Bond Max artifact

2018-10-11 Thread Dr. Jason Rosen via Histonet
Hi Greg,

It has been a problem for months across hundreds of kits. We added some washes
and more dips in the DI water after the run to the protocol. It helps a little
bit. I am going to try a more things in the next few weeks and I will keep the
community updated.

Hi Linda,
Is this a problem that has just started or has the red kit always had this
artefact?

   - If it just started, perhaps it is a problem with a particular kit lot
number (or numbers). If so, get Leica Tech service involved to see if
anyone else has reported a problem.

   - If it has been an ongoing issue for the red kit then perhaps you need
to modify the Red Kit protocol by adding additional (and/or longer) wash
steps. Here again Tech Service can help you with this.
   - The only other thing I can think to ask is: Do you have anyone new
operating the instrument? Could a new person be putting undiluted Bond Wash
   in the bulk container?

Let us know what you find out. Good luck.

Re: [Histonet] Leica Bond Max artifact

2018-10-10 Thread Cathy M. Mathis/Comparative Medicine via Histonet
Hello Dr. Rosen and Histonetters,
We have used a Leica Bond RX for over 2 years now, cleaning and caring for it
just as you do. My work is mostly animal which is why we mainly choose the red
chromogen alkaline phosphatase detection kit. We see the same artifact
sporadically. Sometimes we will see it for several days and then maybe a week
goes by and we don't see any of it. It may only be a small amount and I have
seen it basically all over the slide. I know it is not related to our tissues
or slides. Our Leica rep came out and increased washes but it really doesn't
help. If anyone has a solution to the problem, I would also be interested.
Best to all,
Cathy

Here is a message I am posting for Dr. Rosen:
Hello,

I am a longtime admirer of this list, and I am sure someone on here can help us
out

We have an artifact on our immunohistochemistry (IHC) slides.

I will attempt to attach pictures using the upload tool. In case that doesn?t
work, I am attaching a google drive link to some jpgs. The artifact is usually
golden brown to red/pink in color, is clustered, appears both on top of the
tissue and around it. If we do an aggressive rundown, it appears less red/brown
and bluer.

We are using a Leica Bond MAX IHC system which is about a year old. We usually
do two runs a day and 95% of the time we only use a red detection kit. We only
see this on our red detection kit on our Leica bond. The brown is unaffected.
The corresponding H slides are fine.
We started having this artifact after a month or two of production. 85% of the
time we have some sort of artifact. Sometimes its perfect, sometimes it makes
the tissue unreadable. We haven?t figured out any precipitating factors.

We have tried a number of things, but we still can?t get control of this.
Here is a brief overview of our protocols:
- Processor: Leica ASP300s tissue processor.
-Parrafin = Mercedes HWWX (dual purpose embedding and infiltration parrafin)
melting point 57c ? we tried switching, and it didn?t help.
- We only use distilled water in our baths
- Mercedes Starfrost charged slides
- Oven for 30 min at 62c
- We always use a cold red detection kit
-Sometimes our runs are delayed but not more than 4 hours
- Rundown is done in our stainer (standardized), and slides are coverslipped
with tape.
- The bulk fluid containers in the bond are cleaned weekly and refrigerated
between runs.
- The probe is cleaned every 300 slides.
- Mixing wells are cleaned out every two weeks and replaced often.
-Covertiles are well tended to.

We have tried to correct this problem, but we can't seem to fix it.
Any ideas?

Good Morning,  I have a question about the Leica Bond Max reagents.  Has anyone
ever heard of purchasing the reagents from a 3rd party rather than from Leica ?
If so, what is the company's contact info.

Thanks for your help.

Hi all,

I am researching the Leica Bond Max immunostainer and I like its features as
advertised.

The advertisement however states that the system is completely open, but after
discussing it in length with the Leica rep I am finding that this is actually
not so...

We are a research core and work with mice and rats. Most of the pre-made
antibody are designed towards human tests, so we won't use them. Then we would
still be stuck with lots of money for consumables (as I understood all sold as
kits with the antibodies included). We won't be using any of the antibodies,
only the rest of the solutions... Is there a way out of this trap? Is there
something that I didn't understand?

My question is:
Does anybody use Leica Bond as an open system and use other companies antibody,
buffers and antigen retrieval protocols? If you do, what have you done so far?
(Would you share your protocols?) We like the BioCare products and our helpful
rep :-)
Did anybody purchase the Research dongle? Do we really need it? How did it help
you improve your applications? I was told that depending on what I want
unlocked, we would have to spend $14000 to $27000 extra for it. OUCH!!! This is
not a hospital where we can charge the insurance or patients for this...

I need this info ASAP, because we have to make our decision in a day and there
is no time to demo the unit (nor anything else).
I understand that it is great for pathology, but how about animal research? It
costs A LOT!!! We better know what are we getting ourselves into.

Please help! Any advise and suggestions that I can get would be highly
appreciated.

2009-04-01 Thread Jackie M O'Connor
I agree with Linda. I researched 5 different automated IHC stainers to
be used for toxicology research applications, and found the intelliPATH to
be the best solution for our needs.
Biocare, as well, has a vast array of anti rat and mouse primary
antibodies and detection systems. Additionally, the cost for the
intelliPATH is quite reasonable, compared to the other platforms.

Michelle,

We also only work with animal tissue. Have you considered researching
Biocare's Intellipath immunostainer? We also love their products and this
is an open system. I am not the one who made the purchase decision or
used both of the stainers, but, although the Leica immunostainer was also
well liked, for our purposes (animal tissue), the Intellipath was chosen.
