Hi

If you are using the peroxidase method the hydrogen peroxide step will remove 
the eosin and some of the haematoxylin.

No need to do a decolorisation step before staining.

As Gudren said the only problem may be not having coated slides.

Oh no!!! We cut all slides on + 

BUT you can do this. It’s what we do when we have H on plain slides.

Whether  you have a dry unstain from another lab that sent u a slide, or your 
H that you just took the coverslip off…

Place it in. Coplin jar with 1-3 DROPS of formalin, cover, place In 60ft degree 
oven for 60 min. You want the bottom of the jar to be moist not floating in it 

After 60 min remove and open UNDER A HOOD (it’s a vaporized carcinogen)
Air out 10-15 min… place on stainer 

The drops vaporize and the aldehyde goes through tissue and make it adhere to 
the slide permanently.

I do this when I cut nails or anything that won’t stay on a plus slide.

Hi Nancy,
You should have no problem doing so. If you want proof, just run a couple
of your IHC control sections through the H stainer as you would for a
patient specimen (so right through to coverslipping) and then start the
process of removing coverslip, hydration, destain in acid alcohol and then
run slides on your IHC platform as you would -without the dewax step
obviously. :-)

We do restaining for IHC also, but without destaining at all. Hematoxylin
doesn't matter because nuclei are stained after IHC with hematoxylin anyway.
And Eosin gets lost through all the washing in buffer.
The problem is, that HE section are usually mounted on non-adhesive slides,
and they float away during the procedure.

Nancy, I would say that it can be done in a very particular situation: i.e.: no 
more tissue available, and type of IHC stain. If it is a dual stain, 
pathologist can not obtain the good results. In the lab I worked before, it was 
done, but this technique was not part of the protocol.

Hi Nancy,

We use our Acid alcohol solution from our AFB Stain. It pulls off the Eosin and 
leaves a faded Hematoxylin behind. We counterstain in Hematoxylin anyway so we 
find this protocol effective. Then we remove the deparaffinization step from 
our IHC protocol and run the slide.

1% Acid Alcohol
70% Alcohol..99ml
HCL...1ml
