After struggling with this for some hours I finally gave in, backed up 
my databases, removed MySQL 5.0.15, and re-installed it.


It seems I now have a normal 'superuser' root account again. I still 
have no idea what went wrong. But at least it appears I can get in and 
perform administrative tasks, which is a relief.

Are you talking about root the OS user or root the MySQL user account? 
One or more of the following sections may apply to your situation:

MySQL may have a user called root or not. It all depends on how it has 
been setup.
