A 5-day Workshop, taught by leading experts, presenting the basic concepts of 
pharmacokinetics with pharmaceutical and clinical applications.

For Whom Intended - Pharmaceutical scientists, pharmacologists and other 
scientists, pharmacists, and physicians wishing to understand and apply 
pharmacokinetic principles in drug selection, drug development, protocol 
design, drug evaluation, and drug registration.  Emphasis is placed on relating 
pharmacokinetics to underlying physiological processes, with a brief 
introduction to pharmacodynamics.  Only limited exposure to pharmacokinetics is 
assumed. Primary emphasis on small molecules but consideration also of 
biologics.

The Lecturer / Senior Lecturer role aims to advance research in mechanistic and 
translational pharmacokinetics and pharmacodynamics to complement the

Applications are invited for a Research Associate in PKPD modelling, in the 
Division of Pharmacy and Optometry. The post will be tenable on a fixed term 
basis of 24 months in the first instance.

You will become a member of the Centre of Applied Pharmacokinetic Research 
(CAPKR) and be fully involved in a project entitled 'Development of Integrated 
population PKPD and PBPKPD models' You will hold a PhD in a biomedical, 
mathematics/statistics, engineering, pharmaceutical or systems modelling 
research area, and have a background in pharmacometrics, PKPD or PBPK 
modelling. Previous experience in mixed effects modelling and associated 
software packages as well as working with industrial partners, would be 
desirable. Emphasis will be placed on knowledge of biostatistics and developing 
publication record.

Division of Pharmacy and Optometry. The post will be tenable on a fixed term 
basis of 24 months in the first instance.

You will become a member of the Centre of Applied Pharmacokinetic Research 
(CAPKR) and be fully involved in a project entitled 'Development of Integrated 
population PKPD and PBPKPD models' You will hold a PhD in a biomedical, 
mathematics/statistics, engineering, pharmaceutical or systems modelling 
research area, and have a background in pharmacometrics, PKPD or PBPK 
modelling. Previous experience in mixed effects modelling and associated 
software packages as well as working with industrial partners, would be 
desirable. Emphasis will be placed on knowledge of biostatistics and developing 
publication record.

Exciting PhD opportunity at the University of Manchester funded by BBSRC 
Collaborative Training Partnership, in collaboration with AstraZeneca (Dr 
Meno-Tetang) and University at Buffalo, SUNY (Prof Mager).  The PhD student 
will be charged with developing a QSP/ML platform to study the translational 
potential of animal models of chronic neuroinflammation.  The funding covers UK 
tuition fees and stipend. Although, this only opens to UK applicants,

Last call for applications for September 2020 start on MSc in Model-based Drug 
Development (MBDD) and CPD units for standalone modules.

The MSc programme focuses on the modelling and simulation skills that are 
currently transforming the drug development industry. The course has been 
developed with an emphasis on mechanistic approaches to assessing and 
predicting pharmacokinetics and pharmacodynamics (PKPD), such as 
physiologically-based pharmacokinetics (PBPK). Full-time students benefit from 
immersion in the varied biomedical research environment at The University of 
Manchester, including interaction with research staff at the renowned Centre 
for Applied Pharmacokinetic Research.

We require an UG honours degree (minimum Lower Second or equivalent) from a 
reputable internationally-recognised institution.  You will qualify as a 
modeller with key skills in computational approaches in pharmacokinetics and 
pharmacodynamics. Some of our modules include Introduction to Basic 
Pharmacokinetics and Pharmacodynamics, In-vito-In-vivo extrapolation (IVIVE) in 
ADME,  Physiologically- based Pharmacokinetic Modelling, Advanced topics 
(including modelling of absorption) in PBPK, Data Analysis in Pharmacokinetics 
and Pharmacodynamics and Research project.

The aim of the course is to provide specialist knowledge and skills that are 
highly relevant for a career linked to drug development and pharmaceutical 
industry. On the course you will develop and acquire experience of using 
different software platforms related to pharmacometric data analysis including 
R, Phoenix, NONMEM, MATLAB, Simcyp, and MONOLIX. The course emphasises the 
development of problem-solving skills. A large portion of the learning involves 
structured problems requiring you to apply theory and practical skills to solve 
typical problems that arise in drug development.

Our distance learning option is ideal for scientists linked to the 
pharmaceutical industry who wish to expand their expertise while working in the 
industry. It is also possible to take certain modules/units on the programme as

Applications are invited for the post of Lecturer in Applied Pharmacokinetics 
and Pharmacodynamics who will be based in the Centre for Applied 
Pharmacokinetic Research (CAPKR) in the Division of Pharmacy and Optometry. 
CAPKR is an internationally recognised, academic-led centre of excellence for 
research and training in quantitative prediction of human pharmacokinetics 
(https://www.capkr.manchester.ac.uk/ ).

You will have a first degree either in a biologically related subject (e.g. 
pharmacy, pharmacology or systems biology) or in applied 
mathematics/engineering, with a PhD involving modelling in the area of clinical 
pharmacology. You should also have a strong interest in physiologically-based 
pharmacokinetics, pharmacodynamics, biological molecules or precision medicines 
and will have demonstrated an ability to carry out independent, 
(inter)nationally competitive research. In addition, you should be able to 
demonstrate an upward trajectory in your research terms of quality, as measured 
by publications and contribution to grant income. Evidence of multidisciplinary 
and/or clinical collaboration would be beneficial.

You will be expected to drive forward an independent research programme to 
complement ongoing activities in the CAPKR team and the Division, where there 
is already a strong international track record of work. It is expected that you 
will setup appropriate links with basic science and clinical groups across the 
University in order to initiate projects that will enable advancement of your 
research programme to an internationally competitive/internationally leading 
standard. You will be cognisant of the potential for knowledge transfer in your 
research.

Additionally, you will contribute to teaching in the Division of Pharmacy and 
Optometry that enhance the current undergraduate and postgraduate programmes 
including the MPharm degree and the MSc in Model-Based Drug Development and 
develop teaching and assessment methods and input to curriculum development, as 
required.

Project Description
Cough is the most common reason for which medical advice is sought both at 
primary and secondary health care levels. It has been reported that the 
prevalence of cough in the UK is about 12% and an estimated amount of 2 billion 
dollars is spent on cough and cold medications in a year in the USA. Despite 
the prevalence of cough, understanding of the mechanisms that lead to coughing 
in different diseases is poorly understood; this has made drug development for 
treatment of cough even more difficult. Cough challenge test using inhalation 
agents such as capsaicin has been used to induce and quantify cough with the 
aim of understanding the mechanisms that underline cough and assess efficacy of 
antitussive therapies. Capsaicin cough challenge test has been designed and 
tested by a multi-disciplinary research team (lead by Prof Jacky Smith) at the 
Manchester University NHS Foundation Trust, Wythenshawe.

The objective of this project is to develop mechanistic models for describing 
the data obtained from capsaicin cough challenge test. This model will allow 
better interpretation of the test, provide insight into the mechanisms of cough 
and boosts developments of antitussive therapies. It will also help to refine 
statistical design of the test.

Project Description
Community-acquired pneumonia (CAP) is a leading cause of morbidity and 
mortality in adults and children worldwide1. In the UK, CAP accounts for over 
100,000 hospital admissions annually. Levofloxacin is a broad spectrum 
antibiotic licenced for the treatment of CAP. Resistance of levofloxacin is 
increasingly identified in patients with CAP. The current, fixed dose of 
levofloxacin was established in early phase trials of healthy volunteers and 
non-critically ill patients. Antibiotic pharmacokinetics has been demonstrated 
to be different in critically ill patients compared to healthy volunteers with 
marked between subject variability. Between-subject variability results in 
under-dosing and treatment failure in a significant proportion of patients. The 
optimal dose of levofloxacin in critically ill patients with CAP is still not 
clearly defined.

The objective of this project is to develop a model-based precision dosing 
algorithm for levofloxacin in critically ill patients to improve treatment 
outcome, using mechanistic model that account for between-subject variability 
in the pharmacokinetics.

The Manchester Pharmacy School (MPS) and its Centre for Applied Pharmacokinetic 
Research (CAPKR) invite applications for the position of Senior Lecturer. 
CAPKR’s high volume of research in quantitative prediction of human drug 
metabolism and pharmacokinetics (DMPK) has established MPS as a world-leading 
centre of excellence in this area. Our goal is to advance translation from in 
vitro to in vivo within physiologically-based paradigms and under different 
pathological conditions in order to improve safe use of medicines and provide 
the right dose to the patients. The physiologically-based pharmacokinetic 
(PBPK) models and prediction strategies developed and popularised by our DMPK 
group are now an integral part of drug development and drug regulatory 
submissions.



The Manchester Pharmacy School is seeking to appoint a motivated, dynamic and 
enthusiastic academic with outstanding research skills to develop and deliver 
DMPK research, with a specific focus of expertise in biologics. Involvement in 
precision medicines would also be encouraged. The post holder is expected to 
have an excellent track record of publications and securing research grants. 
The candidate will complement existing MPS strengths and expertise in the areas 
of drug metabolism and transporters, PBPK modelling and pharmacometrics (for

candidate is expected to work closely with colleagues in CAPKR, in addition to 
interdisciplinary work with research institutes elsewhere in the University of

Development of a precision dosing tool for simvastatin treatment in children 
and adolescents with dyslipidaemias

Project Description
Dyslipidaemia is an important etiologic factor in the development of 
cardiovascular disease. Simvastatin is a HMG-CoA reductase inhibitor, commonly 
used to treat lipid disorders and reduce the probability of a cardiovascular 
event. Simvastatin is administered as a lactone and requires conversion to 
simvastatin acid to become therapeutically active. Its complex pharmacokinetics 
is associated with inter-conversion between lactone-acid forms and affected by 
multiple metabolic enzymes (CYP3A4) and transporters (OATP1B1, BCRP).

The objective of this project is to develop a mechanistic population PBPK model 
for simvastatin lactone and acid in children/adolescents with dyslipidaemia. 
The model will allow prediction of simvastatin concentration profiles in liver 
(efficacy) and muscle (toxicity) in this patient population group.

Although widely used in adults, guidance on the individual dosage regimen of 
simvastatin in children and adolescents with dyslipidaemia is lacking. 
Mechanistic model that describes accurately complex simvastatin 
pharmacokinetics in children is essential as a precision dosing tool that would 
also minimise the risk of muscle toxicity (the most serious adverse effect). 
Model development will be based on our published mechanistic simvastatin model 
in adults and will account for physiological differences between populations 
and ontogeny of transporters/enzymes of relevance. Plasma concentrations of 
simvastatin acid and lactone measured in children/young adults will be analysed 
simultaneously with nonlinear mixed effect modelling approach in NONMEM. 
Clinical data will be provided from our collaborator at Children's Mercy 
Hospital and Clinics, University of Missouri, Kansas City. Information on 
patients' age, body mass index, LDL level and OATP1B1 genotype will be 
considered as covariates in the model development. Mevalonate will be measured 
in plasma samples as a biomarker of HMG-CoA reductase inhibition to allow 
comparison of pharmacodynamic effect in children relative to adults. The 
modelling aspects of this project have strong foundations and build upon 
previous and ongoing research in our group.

This 3.5-year full-time MRC DTP studentship provides full support for tuition 
fees, annual tax-free stipend at Research Council UK rates (currently £14, 057) 
and conference/travel allowance. The project is due to commence October 2016 
and is open to UK/EU nationals only due to the nature of the funding.

The successful candidate will receive training in a wide range of research 
skills including quantitative techniques such as modelling and simulation.

Applicants should hold (or be expected to obtain) a minimum upper-second class 
undergraduate degree in a related area. A relevant Masters degree or equivalent

A Research Associate position is available in the Newcastle Cancer Centre 
Clinical Pharmacology Group for a talented PhD-qualified researcher with a 
strong interest in clinical pharmacology and/or drug development.

The post is funded by Cancer Research UK and is available from the 1st November 
2014 for a period of 2 years in the first instance, under the supervision of Dr 
Gareth Veal. This is an excellent career development opportunity for an 
early-career researcher looking to develop interests in clinical pharmacology 
in a cancer setting.

The successful candidate will have experience in cancer research and/or a 
background in pharmacology and will work on multiple projects involving the 
development of assays for the quantification of novel small molecule and 
antibody therapies. They will be required to plan and carry out the development 
and validation of novel assays to facilitate the analysis of clinical trial 
samples. All work will be carried out according to GCLP regulations, in which 
the candidate will ideally have some experience.

Hi Nick,
Thanks a lot for your help. The replacement nmbs.bat works as you described. I 
agree with your comment about the robustness of bootstrap estimates in general, 
but my particular application requires the covariance matrix for each run.
On a related note, as part of development efforts for NONMEM, it would really 
be helpful if any NONMEM estimates which are available in the .lst file can be 
included in a $TABLE file. I realize there may be clever ways to do this using 
abbreviated code but these are usually accessible to more advanced users.

WORKSHOP CONTENT

The Workshop is divided into lectures and small group workshops, in which 
participants will solve practical problems. A manual, comprising lecture 
outlines, derivations, problem sets with answers, and additional reading 
material will be provided. Participants should come with a scientific hand 
calculator. 

 

TOPICS TO BE COVERED INCLUDE:

1.  Physiological aspects of drug absorption, distribution and 
elimination 
2.  Clearance concepts in pharmacokinetics 
3.  Assessment of pharmacokinetic parameters, from plasma and urine 
data, following intravenous bolus, constant rate infusion and extravascular 
(oral) administration 
4.  Assessment of rate and extent of drug absorption; 
bioavailability 
5.  Multiple dosing 
6.  Integration of physiological concepts with pharmacokinetics 
7.  Interspecies scaling 
8.  Distribution kinetics: introduction to assessment of parameters 
and consequences in pharmacokinetics 
9.  Pharmacological Response
