Hello All,
I have purified recombinant Aspergillus CYP51 from E.coli. But it seems that
there is protein there but it is white and not red. No
hemeprotein is present as seen in spectral studies or by eye. The spectral
studies were done ~ 1mg/ml concentration. I did include 5-aminolevulinic acid
Dear Tom,
There is a nice paper which addresses your problem. They coexpressed two
representative Tyr kinases known for autophosphorylation activity with YopH Tyr
phosphatase (Assuming yours is also a Tyr Kinase). You may contact the authors
for the expression plasmid.
High yield bacterial exp
Dear CCPeers
I wanted to thank everyone for sharing their experiences and giving valuable
advice on CYP51 protein expression. It seems that special vectors like pCW-Lic
or pcWORi is needed for efficient expression. I have not found any commercial
source for them though. Could anybody be willing
Hi CCPers
Can anyone provide insights about expressing heme containing proteins in E.col?
Does E.coli need any porphyrin precursor during expression or you need special
E.coli strains. I have references mentioning delta-aminolevulinic acid for one
ortholog but none for another. The enzyme is CY
Hello CCPeers,
I am trying to download CCP4 and Coot on to my new Mac( Mac OSX 10.6.3). On the
downloading site, I am seeing couple of files, which are the ones to be

Are there any other files to be downloaded? Tcl/Tk etc?
