The hyaluronidase is digesting out hyaluronic acid, a component found in connective tissue mucins. This reaction is used to differentiate connective tissue mucins from epithelial mucins. Hyaluronic acid and epithelial sialomucins will give the same staining results, both positive for alcian

Rinse in dist water ( or tap water) x3 over 30 mins No big deal to do this but nice to get rid of extra smells in your processing reagents ( depending on where you start processing: I start in 90% alcoholsure, I ask for recd specimens to be rinsed in water then transferred to 70% alcohol
