I am trying to cut frozen leaf sections at 6 and 8 microns. All I am getting
is bunched up leaf. What is the trick/ I am cutting at -20 should be cutting
at a lower temp and cutting thicker?

We are having difficulty with a particulate set of very, very fatty mouse
livers. The normal livers from this set stay on the slides the fatty livers
fall off. We have used different types of charged slides and we have even
tried to drench the charged slides in Stay-On, dry them and then put

I have a client who wants us to process,embed and cut saggital sections of a
whole mouse which is 8-9 months old.
I have a pretty good idea of what needs to be done but I would like some
feedback from those who have more experience than I do.

We have a client who is interested in studying both fibrin loose in the mouse
tissue as well as intravascular (arterial or venous). It has been suggested I
do either a Picro-Mallory or a Martius, Scarlet Blue (MSB) stain. Is there
any other stains I could do, which is the most popular?
We are having problems getting our CD31 to work well on our NBF fixed mouse
tissue. There is staining but it is very pale.
The CD31 comes from Bio-care and we are are using the IntelliPATH stainer.
We have tried:
- Trypsine 37C 15 min / Trypsine RT 30 min
- Trypsine (1:2 and 1:3)

Hi All,
Over the past year we have had a recurring problems with our HE's. The come
out of our Gemini Stainer stripped but not on every run; we can go for months
without a problem and then all of a sudden it is back again. The stripping is

0012. We have so many back up systems in place that it is almost as if I never

Hi All,
It has come to my attention recently that some of our histology clients are
submitting their samples in PBS after fixation, instead of 70% alcohol. When I
ask them why they don't know and say they have been told to. I have always
followed the procedure that after fixation specimens
We have a new project pending with mouse eyes. The lens must remain intact.
What is the best method to process, embed and cut them? Paraffin or glycol
methacrylate?
