Hello:
Does anyone have a good manual or automatic H&E staining protocol that will
work well on multiple Paraffin embedded tissues?

Obviously, you're going to get a much different result using a
polychromatic stain, than by doing a standard H&E. But if the pathologists
like it...
I'm not even sure if they could call that stain an H&E in their dictation.
Seems like that would open them up to liability if, God forbid, they ever
Has anyone tried using Hologic's Thin Prep solutions (Nuclear stain, rinse,
and bluing) to do their H&E stains?
I am trying to validate a new stainer and in the process the pathologists
want to tweak our protocol to get a better stain and thought maybe we could
try this to see about saving money i
Dear All,
There are many variants to the standard H + E protocol. I'm seeking to
optimize the current protocol our laboratory uses (pasted below). Do you
guys have any suggestions of optimization (e.g. cutting down on the long
water rinse after hematoxylin and increasing bluing reagent time)? Or i
Does anyone have a good H&E stain for bone sections in glycol methacrylate.
There is alginate and collagen in the samples and it is making it very
difficult to read and differentiate between the old host bone and the new bone
or to see the osteoblasts and marrow. My information is listed below
Hi everyone,
I have a colleague that has been asked to do H&E staining on a frozen specimen
collected on tape. The tape has been attached to the slide using nail polish.
Can't use the CryoJane system due to the light flash. Her question is "will the
nail polish disintegrate during the immersion

Hi Charles,
If my memory serves, Clarifier 1 and Clarifier 2 are different mostly in that
one is an alcoholic solution and the other an aqueous one. So they both behave
differently in their mechanism of action. The alcohol based reagent should
remove more hematoxylin from the cells than the aqu
Usually inconsistency in H&E staining has two different sources: either the
staining process itself, or the previous tissue processing sequence.Your
staining protocol has a "standard" sequence but I would add an additional
xylene and would increase the time in hemtoxylin to 7 minutes for you wil

Hello all,
We've been running into an issue lately with our H&E staining. The
hematoxylin and eosin are light on and off but the chromatin is
consistently staining light. I have changed the time in hematoxylin to
longer, used a fresh lot of both hematoxylin and our clarifier solution and

Was wondering if anyone else is experiencing any h&e staining issues recently
using the symphony stainer? Our lab has been experiencing uneven staining and
dark mucin staining especially on our Gi biopsies. This has been happening on
specimens that have been processed on different processors, tw
I am interested in the answer as I use this same technique on my rodent
samples and have not had this issue.

We had some rat tissues that were removed at necropsy and immediately fixed in
formalin for 48 hrs. Since the tissues could not be processed after the 48 hr
fixation they were transferred to 70% ETOH for about a week, then processed and
stained with H&E.
The H&E staining looks

Good morning Histonetters,
Can anyone tell me if you are using a particular company's reagents for your
H&E staining that are pre-made and meant to work together? Also, is the cost
significantly more than buying separate reagents and making up your own?
Thanks,

Hello,
I was wondering if those of you who are using H & E stainers with heating
capabilities/ovens could tell me what your average run time is. What instrument
are you using? and is the length of time affected by the number of racks being
stained?
Thank you,
Toni Rathborne
