On many filesystems, that is true. Not all, though.
Also, MIX is a nice compromise between mbox and maildir, since it can keep
multiple messages per file
with something UW-ish, you
should be looking at that.
Even if you go to UW imap, you should expect to have to convert to some
other format (likely MIX).
If you want to stick with traditional mailbox formats and you need to
support really large folders, you should investigate dovecot.

Is it possible to access mix mailboxes this way or can they only be read via
imap? We're looking for something that is like opening an mbox file using
Local Folders on Thunderbird.

our campus had deployed some kind of decent
digital drop box software and then trained our customers in how to use
it. It's too late for that now; people have already become accustomed to
using email as the sole method for digital file distribution.
