I have a question regarding MailMate signature behavior. My signature
setting is as follows,
Default signature placement: Top
Caret placement: Above Signature
Everything works as expected as long as the signature has been inserted
before writing anything into the new email window.

If you mean the sounds then I think you are correct. I think the “Do
Not Disturb” setting only affects the notifications displayed.
Both the sound and the counter.
For now, you can disable sounds in the Counters preferences pane of
MailMate.
