You have to change it into a region - it is not enough that the polyline
looks like a region.

If you have MapInfo 7.0 you can split an object with a polyline (Objects >
Polyline split...)
If you have an older version of MapInfo you need a polygon - not just a
closed polyline (Objects > Split).
In both cases you have to select the linie to split, make sure the layer is
editable.

Select the polyline, Set Target On
Draw a polyline that cuts across the targeted polyline (you may want to use object
snap to snap a node of the polyline to the exact point where you want to divide the
