No for UDP protocol("udp") and same IP:port should be used, why ?
Most NATs are "port restricted cone" NAT, so they won't allow traffic in
from the end point to where you haven't send data.

Following is the scenario on which my question is based:
1> UAC-1 sends a REGISTER request using UDP from source port 20,000 to UAS-2 at
destination port 5060.
2> UAS-2 responds back with a 200 OK to above request from UDP source port
30,000 to destination port 20,000.
