How can I ensure that James use a specific interface to send all outgoing message through?
Outgoing messages for "Instance A" must use eth0 and outgoing messages for "Instance B" must use eth0:1.

Check out the options for RemoteDelivery mailet. Sounds like you want the <bind> parameter.
