Yes.  This is documented in the manual.  I'd recommend reading:
http://tcpreplay.synfin.net/trac/wiki/usage#RewritingIPsforDual-NIC
but there are other ways of doing this as well (pseudo-NAT for
example).

Not a problem.  If you're planning on looping these files (playing
them multiple times in the same session) I would suggest compiling
with --enable-64bits so that the counters/statistics can deal with
values > 2^32.
