You can, but if you reject at SMTP time and don't store the message, you can't provide an easy link or reply-to address for the sender to release his message with.
Yes, an SMTP-level C/R system would need to store the message, but it would not need to send a separate challenge/bounce e-mail -- the upstream SMTP server would handle that. This can be accomplished with qmail and something like sa-analyze.
