I suspect the the admins of locamotion.org could give you more
extensive guidance, but they are probably visiting with their families
at the present time in celebration of the Christmas holidays.  I am
just a fellow Pootle fan and user.

As a start, I would like to point you to

Hopefully this page from the Translation Toolkit (which is developed
by the Pootle and Virtaal Developers, which in many ways represents an
extension of and a scriptable set of the basic functions they employ
in those programs.  I hope that reading that page and experimenting
some with that function may answer some (or even all) of your
questions, but in any event, it should serve to clarify some of the
available tools, if not exactly the workflow that is used on
pootle.locamotion.org for the African languages work on Mozilla
efforts that they host.
