@Will no clue what the difference might be. Try to export userroles workspace and compare roles permissions between instance where it is working and where not. Also as Richard suggested, try to upgrade to 4.4.8.
@Andreas Difference might be that while in 4.4.x and older versions read access to the root of the workspace was always granted and you could not deny it, it is actually possible to deny complete access to the workspace for user in 4.5. As in Wills case I would suggest upgrade to the latest release - 4.5.8 in this case. For both of you - If the above doesn't help please paste complete stacktrace of the execption generated in the logs to pastie.org or similar service and put link to it here.
